肌间刺缺失对斑马鱼肌肉发育的影响

【摘要】 研究表明,肌间刺在肌肉支撑和力量传递方面发挥着一定的作用。为了验证这一假设,本文利用肌间刺部分缺失表现型从肌肉特异性基因表达和肌纤维密度两方面分析了肌间刺缺失对肌肉生长发育的影响。基因表达方面,比较分析了斑马鱼背鳍前部肌肉和背鳍后部肌肉myod,myog,myf5,mef2和sox6 5个基因在15dpf,30dpf,45dpf,60dpf和75dpf的表达。结果显示:同一时期、相同部位,与野生型相比,肌间刺突变体斑马鱼5个肌肉组织基因的表达无明显差异;相同部位、单个基因的表达随着时间的推移,其表达活性也逐渐降低;相同生长时期,单个基因背鳍前后肌肉组织的表达量也相差不大。此外,对比观察了背鳍前后部单位面积肌纤维数目在各发育时期的变化,相同时期相同部位肌间刺突变型斑马鱼单位面积内肌纤维数目与野生型斑马鱼没有明显差异;相同部位单位面积内的肌纤维数随生长时间的推移而逐渐减少;相同发育时期背鳍前后单位面积肌纤维数目关系背鳍后部多于背鳍前部。
【Abstract】 Intermuscular-bones(Imbs) were thought to function as supporting muscles and transmitting strength. To test this hypothesis, we used an Imbs partial deletion fish lineage to analyze whether Imbs’ deletion impacted muscle’s development and growth from two aspects, expression of muscle-specific gene and density of muscle fiber. Five muscle-specific genes including myod, myog, myf5, mef2 and sox6, were selected to test and verify expressional differences among 15dph(days post hatch), 30 dph, 45 dph, 60 dph and 75 dph in anterior and posterior muscle of dorsal fin. Compared to the wild-type zebrafish, the mutants showed no significant differences of the expressions of the 5 genes. Among of the different development stages, the expression profiles of 5 genes in the mutant were similar to that in wild-type zebrafish. In addition, the expression levels of the 5 genes in anterior muscle to dorsal fin were close to posterior ones’. Furthermore, we carried out the histological analyses of muscle fiber in anterior and posterior dorsal fin at different development stages. The results showed there were no significant differences of muscle fibers’ density in the same period both at anterior and posterior muscle of dorsal fin. The density of muscle fiber declined gradually over time. Moreover, the density of anterior part was more than posterior part. In conclusion, the lackness of intermuscular-bones has no substantial influence on muscle’s development.
