斑节对虾新型高血糖激素基因的特异性表达及组织定位分析

【摘要】 甲壳动物高血糖激素家族是眼柄中参与神经内分泌调控最重要的家族之一,但是有证据表明,眼柄神经节并非该家族激素唯一的合成场所。实验获得一种斑节对虾新型高血糖激素基因(PmH HLP),其全长cDNA包含354bp的开放阅读框,推测编码118Aa多肽,序列含有CHH-I型基因的基本结构:信号肽、CPRP区和成熟肽。Pm HHLP与两种中国明对虾高血糖激素有67%和63%的相似性,在进化上与两者独立聚为一支。该基因在在卵巢中有微弱表达,在精荚和输精管表达量较高。利用20天精荚再生得到的精荚和输精管样品进行荧光定量和荧光原位杂交,结果表明PmH HLP的表达量随精荚成熟而升高,但与精子数量关系不显著;其定位于精荚和输精管的内柱状上皮中,随着精荚的成熟迅速增强,而在输精管中则相对稳定。综合上述结果表明,PmH HLP参与了精荚囊的形成,可能调控精荚的发育和成熟,与已知的高血糖激素大不相同。
【Abstract】 Crustacean hyperglycemic hormones(CHHs) are traditionally considered as a neuroendocrine hormone family secreted from eyestalks. However, a newly found CHH gene named Pm HHLP could not be detected in the eyestalks of Penaeus monodon. The full-length cD NA of PmH HLP was 354 bp, predicting a precursor of 118 residues. PmH HLP contain a CHH precursor-related peptide(CPRP) region which are characteristic of the CHH-I peptides, a signal peptide and a mature peptide with six highly conserved cysteine residues inside. Its mature peptide shared 67% and 63% identities with another two CHH-like genes in Fenneropenaeus chinensis, belonging to an independent branch. Its transcripts were detected specifically in the spermatophore sacs and vas deferens of mature male tiger shrimps, and slight in the ovary of the female, while they could not be found in the eyestalks. They were also observed during the 20-days’ regeneration of spermatophore sac of the male shrimps. The expression level of PmH HLP was found rising as the growth of the spermatophore sacs, and showed little significance with the quantity of the sperm. The FISH probe of PmH HLP was located in the inner epithelium of the both tissues, probably in the inner epithelial cells. Its signal increased and flowed around as the layer of the spermatophore sacs thickening, while it was stable in vas deferens. To sum up, PmH HLP is believed involved in the forming of the spermatophore sacs, and may be in the regulation of the maturation of them, which have out of our knowledge in the CHHs.
