黄姑鱼锰超氧化物歧化酶的克隆及其对氨氮和亚硝酸氮胁迫的表达分析

【摘要】 锰超氧化物歧化酶(MnS OD)是一种催化超氧自由基为普通分子氧及过氧化氢的抗氧化酶。本研究中,黄姑鱼MnS OD基因被克隆出来,其全长为949bp,包括38bp的5’、233bp的3’端非编码区和678bp的开放阅读框,编码225个氨基酸。Real-time PCR分析表明MnS OD在所检测的黄姑鱼各组织中均有表达,并且在心脏中表达量最高。此外,在受到氨氮和亚硝酸氮后,黄姑鱼表现出对氨氮刺激更为敏感,其氨氮和亚硝酸氮的96小时半致死浓度分别为19.63mg/L(或者非离子氨0.85mg/L)和107.65 mg/L,并且其肝脏、鳃、和头肾中MnS OD基因的表达水平明显上调。这些结果可能有助于了解Mn SOD在鱼类中的重要作用。
【Abstract】 Manganese superoxide dismutase(MnS OD) is an antioxidant enzyme catalyzing the superoxide radicals into either ordinary molecular oxygen(O2) or hydrogen peroxide(H2 O2). In this study, the cD NA of MnS OD, cloned and characterized from nibea albiflora, was 949 bp in length including 5’-untranslated region(UTR) of 38 bp, 3? UTR of 233 bp and an ORF of 678 bp encoding a polypeptide of 225 amino acids. The mR NA transcripts of Mn SOD were detected in all examined tissues and the predominant distribution was heart. In addition, when exposured to ammonia or nitrite, nibea albiflora showed lower tolerance to ammonia with the 96 h LC50 of 19.63 mg/L for ammonia nitrogen(or 0.85 mg/L for un-ionized ammonia) and 107.65 mg/L for nitrite nitrogen. Furthermore, the temporal expression of MnS OD was significantly up-regulated in the liver, gill and head-kidney. These results may contribute to better understanding of the roles that MnS OD play in fish.
