布氏鲳鲹GHR1和GHR2基因的克隆及温度和饥饿对其表达的影响

【摘要】 为研究温度和饥饿对生长激素受体的影响,本实验从布氏鲳鲹肝脏中克隆得到两种生长激素受体基因GHR1和GHR2,其cD NA全长分别为2334 bp(编码635个氨基酸)和2248 bp(编码585个氨基酸)。与其他脊椎动物相同,布氏鲳鲹GHR1和GHR2基因存在GHR配体结合区FGEFS,Box1和Box2序列框,但含有不同的半胱氨酸残基数和络氨酸残基数。通过实时荧光定量PCR分析,结果表明,布氏鲳鯵GHR1和GHR2基因在肝脏中表达量最高,肌肉、脂肪、性腺次之。通过对布氏鲳鯵进行不同温度作用实验,结果表明水温为21℃时,布氏鲳鲹肝脏中GH与GHR1基因表达量最高,而水温为33℃时,GHR2基因表达量最高。对布氏鲳鯵进行饥饿复投实验发现,在饥饿第一天,与对照(投喂)组相比,布氏鲳鲹肝脏中GHR1基因表达量显著减少(P<0.05),而GHR2基因表达量显著增加(P<0.05)。在饥饿第三天,布氏鲳鲹GHR1和GHR2基因的表达量均增加。在复投第一天,布氏鲳鲹GHR2基因的表达量与对照组无显著性差异,而GHR1基因的表达量在饥饿第五天与对照组相同。结果表明布氏鲳鲹GHR1和GHR2基因可能对温度和饥饿刺激存在不同的应激机制。

【Abstract】 To clarify the influences of temperature and starvation on growth hormone receptor(GHR), two cD NAs encoding GHR were isolated from the liver of Snubnose pompano(Trachinotus blochii). The two cD NAs were 2,334 bp(635 aa) and 2,248 bp(585 aa) in length and named GHR1 and GHR2, respectively. Similar to other vertebrates, Snubnose pompano GHR1 and GHR2 had GHR ligand binding motif FGEFS regions, Box1 and Box2 regions, but contained different cysteine residues and tyrosine residues. Real-time PCR revealed that Snubnose pompano GHR1 and GHR2 were highly expressed in liver, followed by muscle, adipose, gonad. The effects of different temperature on Snubnose pompano revealed that the GH and GHR1 expressions were the highest in liver at 21°C, while GHR2 expression was the highest at 33°C. Following the fasting and refeeding on Snubnose pompano, compared to the control(fed) group, GHR1 expression in liver significantly decreased on day one(p<0.05) and increased on day three, whereas GHR2 expression was significantly up-regulated on day one, three and five(p<0.05). No significance in Snubnose pompano GHR2 expression was found on the first day of refeeding, and GHR1 expression did not change on day five. These results indicated that Snubnose pompano GHR1 and GHR2 may have different mechanisms for temperature and fasting.
