基于COI序列的日本鲭种群遗传结构与演化历史分析
Population genetic structure and evolutionary history analysis of Scomber japonicus base on the COI gene

【摘要】 为了更合理有效利用中国沿海日本鲭资源,本研究采用线粒体COI基因对来自中国东海(ES-1~ES-7)、台湾海峡(TW)、山东威海(SD)、北太平洋(BT)和西非(XF)共11个日本鲭的种群进行了分析。结果显示,290条线粒体COI序列(615bp)共定义27个单倍型,34个变异位点(5.53%),具有较高的单倍型多样性(0.61396±0.02588)和较低核苷酸多样性(0.00196±0.00043),11个群体内的遗传距离值为0.0008~0.0027,A+T(50.2%)的含量与G+C(49.8%)含量相差不大。分子方差分析表明日本鲭的变异主要来源于XF群体与其他10个群体间。AMOVA遗传变异方差分析可以发现,去除XF群体,变异主要来源于群体内。种群历史动态分析表明在13.65万年前的第四纪更新世经历过种群扩张事件。本研究将为太平洋日本鲭渔业资源管理和可持续发展提供科学参考依据。
【Abstract】 In order to take advantage of Scomber japonicus resources along coast of china more efficiently,base on the COI gene, we analyzed the population genetic structure of 11 groups which were collected from the East China Sea(ES-1 ~ ES-7), Taiwan Strait(TW), Shandong Weihai(SD), North Pacific(BT) and West Africa(XF). A total of 27 haplotypes were defined through 290 COI sequences(615bp) and 34 variation sites(5.53%) had been detected. The 11 populations were with high haplotype diversity(0.61396 + 0.02588) and low nucleotide diversity(0.00196 + 0.00043). The genetic distance of 11 populations in value from 0.0008 to 0.0027 and the value of A+T(50.2%) and G+C(49.8%) was similar. Molecular variance and AMOVA genetic variance analysis showed that the variation of Scomber japonicus mainly derived from XF group and other 10 groups. It had experienced population expansion events in the Pleistocene(136-500 Mya). This study will provide scientific references for resource management and sustainable development of Scomber japonicus fishery.
