黄河鲤IL17基因的全基因组鉴定及其对嗜水气单胞菌的响应表达
Genome wide identification of interleukin-17(IL-17) in common carp(Cyprinus carpio) and their expression following Aeromonas hydrophila infection

【摘要】 白细胞介素17(IL17)是一类重要的促炎症性细胞因子,是粘膜免疫应答的重要介质。然而,大多数对IL17基因的研究都集中在哺乳动物上,缺少鱼类物种中的研究。本研究首次从基因组和转录组数据库中表征到了7个鲤鱼(Cyprinus carpio)IL17基因。系统发育分析表明,大多数IL17是高度保守的,且分为三大类。早期发育阶段和健康组织中IL17的表达模式分析显示IL17基因在发育过程的早期是普遍表达的,且在脾、肝、脑、肾中中有较高的表达水平。嗜水气单胞菌在鳃中的响应表达结果显示大多数IL17基因感染后4小时呈现上调趋势,之后逐渐下降,而IL-17A/F2和IL-17N是在感染后的12h呈现上调且L17D2则保持上调趋势,这与IL17D1正好相反。本研究为理解鲤鱼IL17基因的进化,鱼类疾病管理和免疫调节方面提供了重要的基因组资源。
【Abstract】 Interleukin-17(IL-17) family cytokines are known to have pro-inflammatory actions as important mediators of mucosal immune responses. However, most studies of IL-17 genes have focused on mammals, and much less is known of IL-17 genes in fish species. For the first time, seven IL17 gene homologues from common carp were characterized from genomic and transcriptomic databases.Phylogenetic analysis revealed that most of the IL-17 are highly conserved, which could be classified into three subclasses. The expression patterns of IL-17 genes in common carp were examined during early developmental stages and in various healthy tissues, showing that most IL-17 genes were ubiquitously expressed during early development and existed particular tissue-specific in various healthy tissues with a relatively high level in spleen, liver, and kindey. The response expression of Aeromonas hydrophila in the gills showed most IL-17 genes were up-regulated at 4 h post infection and declined gradually, while IL-17A/F2 and IL-17 N were generally up-regulated at 12 h post infection, and IL-17D2 maintained a rising tendency, which is exactly the opposite of IL17D1. Moreover, the whole set of common carp IL-17 gene family research provides a genomic resource for future studies on IL-17 gene evolution, fish disease management and immune regulation.
