南设得兰群岛南极磷虾mtDNA ND6遗传多样性及遗传结构分析
Genetic diversity and structure of Euphausia superba in South Shetland Islands using the mitochondrial ND6 gene

【摘要】 本文采用线粒体ND6基因序列对南设得兰群岛12个站点共493个南极磷虾的遗传多样性,遗传结构和动态历史进行了分析。序列分析表明:南极磷虾ND6基因全长为522bp;碱基组成为T(44.7%),C(16.7%),A(27.3%)和G(11.3%);序列间共存在292个单倍型;序列的核苷酸多样性和单倍型多样性分别为0.01399和0.9803。12个采样站位内和站位间遗传距离接近(在0.013-0.016之间)。分子方差分析表明,遗传变异主要来自于群体内(群体内变异100.21%);同时,基因流和遗传分化指数也表明南极磷虾在南设得兰群岛缺乏一定的遗传结构。群体动态历史研究表明:种群扩张发生在0.0413百万年前的,正处于更新世晚期。该研究不仅加深了对南极磷虾遗传背景的了解,而且为南极磷虾资源的开发,南极生物多样性的保护及生态平衡的维持提供更多的科学信息。
【Abstract】 Using ND6 gene, we surveyed the genetic variation, population structure and demographic history of 493 Euphausia superba from 12 sites in Southern Shetland Islands. The complete ND6 gene of Euphausia superba is 522 bp; the average base composition is T(44.7%), C(16.7%), A(27.3%) and G( 11.3%), respectively; a total of 292 haplotypes were detected; the nucleotide diversity(π) and haplotype diversity(Hd) is 0.00196 ± 0.00043 and 0.61396 ± 0.02588, respectively. Genetic distances within and between 12 sites were very similar(varied from 0.013 to 0.016). The AMOVE analysis suggest that genetic variation may come from within sites; meanwhile Fst and Nm also suggest that Euphausia superba lack of genetic structure in South Shetland Islands(genetic variation within sites 100.21%). Demographic history analysis suggest the expansion occurred in about 0.0413 million years ago, is in the late Pleistocene. Our study not only broaden our comprehending of genetic background of Euphausia superba, but also provide more scientific information on Euphausia superba resources exploitation, biodiversity conservation and ecological balance maintenance in Antarctic.
