不同周龄对NOD背景小鼠胚胎生产效率的影响

【摘要】 目的提高用于进行显微注射的NOD背景小鼠胚胎的生产效率。方法使用不同周龄的NOD背景雌鼠进行实验,分别为4周龄、6周龄、8周龄和10周龄雌鼠,与NOD背景雄鼠进行体外受精,第二天统计每只雌鼠可用胚胎数量,重复两次实验。然后选择效率高的组别进行显微注射,生产转基因小鼠,统计后代情况。结果 10周龄雌鼠体外受精得到的二细胞数量最多,体外受精效果最好。使用10周龄雌鼠体外受精获得的胚胎进行显微注射,8次实验全部获得阳性后代。结论 10周龄NOD背景雌鼠体外受精效果最好,显微注射后可以得到阳性后代。

【Abstract】 Objective To improve the productive efficiency of embryos of NOD mice for microinjection. Methods NOD female mices were divided into four groups: 4 weeks old, 6 weeks old, 8 weeks old and 10 weeks old female, respectively. Male mices were subjected to in vitro fertilization. The next day, the number of embryos per adult female was counted and repeated twice. And then select the high efficiency of the group for microinjection producing transgenic mice, then collect statistics of future generations. Results The number of two-cells embryo in vitro fertilized by 10 week old female mice was the highest, and the effect of in vitro fertilization was the best. Microinjections were performed using embryos obtained from 10-week-old female mices in vitro fertilization, and all of them were positive for all eight trials. Conclusion Embryos obtained from NOD background female in vitro fertilization were microinjected and were more efficient at about 10 weeks of age.
