黄芪总皂苷对H22荷瘤小鼠的肿瘤生长及免疫功能的影响

【摘要】 目的探讨不同浓度的黄芪总皂苷对小鼠肝癌H22肿瘤细胞生长及免疫功能的的影响。方法建立小鼠肝癌H22移植瘤模型,随机分为模型对照组(M)、阳性对照组(CTX)、黄芪总皂苷高、中、低剂量组(G、Z、D),另设空白对照组(K)。称重法检测抑瘤率、脾脏指数、胸腺指数,MTT法检测淋巴细胞的增殖能力及T淋巴细胞的转化能力,ELISA法检测IL-2的含量。结果与空白对照组比较,模型组小鼠的体质量[(24.90±2.82)]明显减轻,胸脾指数[分别为(1.47±0.45)、(3.97±0.05)]显著下降,淋巴细胞的增殖能力及转化能力[分别为(0.10±0.02)、(1.08±0.03)]明显降低,血清IL-2的含量[(87.59±28.60)pg/m1]明显下降;与模型组比较,黄芪总皂苷各干预组可显著抑制模型小鼠体内肿瘤生长。同时,黄芪总皂苷各干预组小鼠的胸脾指数[分别为(2.18±0.34/1.14±0.07)、(2.47±0.54/6.13±0.71)、(2.51±0.46/6.46±0.85)]升高,淋巴细胞增殖能力和T淋巴细胞转化能力[分别为(0.17±0.01/5.42±0.84)、(0.18±0.03/1.15±0.06)、(0.19±0.01/1.20±0.07)]增强,血清IL-2的含量[分别为(117.67±30.28)、(121.80±34.04)、(124.93±37.10)pg/ml]升高。结论黄芪总皂苷可抑制小鼠肝癌H22肿瘤细胞的增殖,其机制可能与增强机体的免疫功能有关。
【Abstract】 Objective To investigate the effect of different concentration of total astragalus saponins on the growth of tumor cell and immune function in H22-bearing mice.Methods The H22 liver tumor model was established,they were divided into tumor bearing group,positive control group,total astragalus saponins low,middle and high dose group.Blank group was also set up.All mice were sacrificed for calculating the tumor inhibition ratios,the indexes of spleen and thymus.The multiplication capacity of lymphocyte and the transformation ability of T lymphocyte was checked by MTT.Meanwhile,the IL-2 content were detected by ELISA.Results Compared with the blank control group,weightbody(24.90±2.82) thymus(1.47±0.45 mg/g) and spleen index(3.97±0.05 mg/g) decreased significantly in the model group(P < 0.05).At the same time,the lymphocyte multiplication capacity(0.10±0.02),the T lymphocyte transformation ability(1.08±0.03),the IL-2 content(87.59±28.60 pg/ml) were obviously downed in the model group(P<0.01).Experiment tests showed that the tumor inhibiting rate of intervention group was better than that that in the model group(P<0.01).Meanwhile,the thymus and spleen indexes in intervention group(2.18±0.34/1.14±0.07、2.47±0.54/6.13±0.71、2.51±0.46/6.46±0.85 mg/g) were higher than that in the model group(P<0.01).The capacity of lymphocyte multiplication and T lymphocyte transformation in intervention group(0.17±0.01/5.42±0.84、0.18±0.03/1.15±0.06、0.19±0.01/1.20±0.07) were higher than that in the model group(P<0.01).Simultaneously,the IL-2 content in the intervention group(117.67±30.28、 121.80±34.04、 124.93±37.10 pg/ml) showed higher than model group(P<0.05 or P<0.01).Conclusion The total astragalus saponin could inhibit the proliferation of tumor cell of H22-bearing mice,The mechanism may be related to enhancing immune function.
