【摘要】 目的研究丹参酮ⅡA对乳腺癌T47D细胞增殖的影响及其可能的分子作用机制。方法正常T47D细胞和应用GPER SiRNA转染构建GPER基因沉默的T47D细胞,分别采用MTT法和流式细胞技术测定不同剂量丹参酮ⅡA对T47D细胞增殖速率和增殖周期分布的影响。采用Western blot方法测定丹参酮ⅡA对T47D细胞中的G蛋白偶联雌激素受体（G protein-coupled estrogen receptor,GPER）、磷酸化细胞外信号调节激酶（p-ERK）、细胞周期蛋白（Cyclin）D表达情况的影响。结果 1×10^-8mol/L雌二醇能够明显促进乳腺癌鳞癌T47D细胞增殖,加入ER受体拮抗剂ICI182 780促增殖作用受到明显抑制。而5×10^-5 mol/L、1×10^-5mol/L、5×10^-6mol/L和1×10^-6mol/L丹参酮ⅡA能够显著抑制乳腺癌鳞癌T47D细胞增殖,该抑制作用可被ER拮抗剂ICI182 780增强。丹参酮ⅡA作用于GPER基因沉默的T47D细胞,该细胞表现出更为明显的生长抑制效应。Western blot检测结果显示5×10^-5mol/L、1×10^-5mol/L、5×10^-6mol/L和1×10^-6 mol/L丹参酮ⅡA可使T47D细胞中GPER、p-ERK、Cyclin D蛋白表达水平显著降低。结论丹参酮ⅡA具有抑制乳腺癌鳞癌T47D细胞增殖的作用,且该抑制效应是通过靶细胞雌激素受体途径实现的;并通过降低GPER、pERK、cyclinD表达量发挥抗增殖作用。更多还原

【Abstract】 Objective:To study the effect of tanshinone ⅡA on proliferation of breast cancer T47 D cells and its possible molecular mechanism.Methods:The effects of different doses of tanshinone ⅡA on proliferation of T47 D cells and GPER gene silencing T47 D cells were measured by MTT assay and flow cytometry analysis.The effects of tanshinone ⅡA on expression levels of G protein-coupled estrogen receptor（GPER）,phospho-extracellular regulate kinase（p-ERK） and cyclinD in T47 D cells were measured by Western blot.Results:1×10^-8 mol/L Estradiol can obviously promote the T47 D breast cancer cell proliferation and such effect could be attenuated by ER antagonist ICI182 780.5×10^-5 mol/L,1×10^-5 mol/L,5×10^-6 mol/L and1×10^-6 mol/L tanshinone ⅡA significantly inhibitT47 D cell proliferation and such effect could be enhanced by ER antagonist ICI182 780.5×10^-5 mol/L,1×10^-5 mol/L,5×10^-6 mol/L and 1×10^-6 mol/L tanshinone ⅡA significantly inhibit T47 D cell proliferation.更多还原
