Enhanced triterpene acid production by Ganoderma lucidum using a feeding stimulus with integration of a two-stage pH control strategy

【摘要】 BACKGROUND:Triterpenes(especially triterpene acids) is one of the major physiologically active components of Ganoderma lucidum.However,submerged fermentation techniques of triterpenes is not optimized for commercial use,as total triterpene yield is still very low.Our aims are to develop a two-stage p H control strategy integrated with a stimulus,an ether extract(EECM,from the medicinal insect,Catharsius molossus),for enhanced G.lucidum triterpene acid production.RESULTS:The triterpene acid production of 302.76 mg/L was obtained in the G.lucidum culture with EECM,which was higher by 60% compared with control cultures not fed EECM.Based on the feeding EECM method,a new two-stage p H control strategy was integrated resulting in triterpene acid yields of 361.47 mg/L.Subsequent studies optimized interactions between EECM feeding and two-stage p H control strategies,which resulted in an optimal EECM concentration of 185.89 mg/L,optimal p H values of 5.07 in the first 72 h incubation and 4.37 after 72 h with a maximum triterpene acid concentration of 449.37 mg/L.This represented a 137% increase compared to that of the control cultures.Furthermore,using a 150-L fermenter under these conditions,a triterpene acid concentration of 431.83 mg/L was achieved.CONCLUSIONS:Enhanced production of triterpene acid in submerged cultures of G.lucidum was achieved using an EECM feeding stimulus integrated with a two-stage p H control strategy in a large scale.
