柱前在线衍生-HPLC法测定发芽糙米中γ-氨基丁酸含量

【摘要】 糙米发芽是糙米在一定温湿度下,胚芽生长形成多元内源酶复合作用的过程。在发芽过程中糙米营养品质、功能特性和食用品质均产生显著性变化,糙米的消化吸收性、口感均得到改善。发芽过程中生成的GABA是一种对用脑人群有益的神经递质。目的:建立柱前在线衍生-高效液相色谱法(HPLC)测定发芽糙米中γ-氨基丁酸(GABA)含量的方法。方法:样品以1:5的料液比、超声功率(占总功率)60%、30℃下超声30min,提取2次。衍生试剂OPA与样品在线混合10次、衍生2min,用Agilent Eclipse Plus C18柱(4.6mm×100mm,3.5μm)分离,流速1nl/min,检测波长338 nm,柱温40℃。结果:GABA在550mg/L范围内线性关系良好,相关系数大于0.9990;最低检出限0.33mg/L,最低定量限1mg/L,平行样品的相对标准偏差(RSD)0.39%2.35%,相同样品6d内测定的RSD值为2.32%,样品测定的回收率94.80%98.40%,RSD为1.82%2.58%。结论:该方法简单快速、灵敏度高、精确,适宜发芽糙米中GABA的检测。

【Abstract】 The germination of brown rice is acombined interaction of multi-endogenous enzymes formed by the growth of embryonic tissues under a certain temperature and humidity condition.During the germination,the nutritional,functional and edible properties of brown rice are significandy changed,as well as the digestibility,absorbability and mouth feel of brown rice products are improved.Among the enhanced nutrients,GABA,as a neurotransmitter,is benefit to human’s brain.Objective;To establish a HPLC method with online precolum derivatization for the determination of "/-amino butyric acid(GABA)in germinated brown rice.Method;the germinated brown rice samples were extracted two times under a certain experimenmtal condition,as the material to liquid ratio was 1:5,ultrasonic power(total power) was 60%,ultrasonic temperature was30℃,and ultrasound time 30 min.The samples were then mixed 10 times with the orthophthalaldehyde(OPA) derived reagents,followed by 2min online pre-column derivation.Samples were separated by HPLC with an Agilent Eclipse Plus C18 column(4.6mm × 100 mm,3.5μm) the parameters were setted as the flow rate 1.OmL/min,the wave length338 nm and the colomn temperature 40℃.Results;Good linearity was observed in the range of 5 50mg/L with the correlation coefficient >0.9990.The detection limit,quantitative detect limit and the relative standard deviation(RSD) among different parallels were 0.33mg/L,1mg/L,and 0.39% 2.35%,respectively.Moreover,the RSD of the same sample at different times within 6d was 2.32%,recovery range was 94.80%98.40%,with RSD 1.82%2.58%.Conclusion;The proposed method was simple,rapid,sensitive,and accurate,which is suitable for the detection of GABA in germinated brown rice.
