白酒中血管紧张素转换酶抑制肽的发现与研究

【摘要】 本研究首次在中国白酒中发现一种多肽,该多肽具有明显的抑制ACE酶活性的作用。采用直接浓缩手段从古井浓香型白酒和国井芝麻香型白酒中分离发现,采用液相色谱-飞行时间质谱联用仪(LC-ESIMS-QTOF)和合成标准品对其氨基酸序列进行了鉴定,其氨基酸序列为Pro-His-Pro(PHP);对其体外抑制血管紧张素转换酶(ACE)的活性测定方法进行了改进,同时对其抑制ACE的半抑制浓度IC50与抑制作用类型进行了测定和研究。IC50为446 mM,PHP和底物HHL能够同时与ACE酶结合,是ACE酶的非竞争性抑制剂。
【Abstract】 The Chinese baijiu samples were concentrated with rotary evaporator,and a peptide with novel ACE inhibitory activity was discovered in the concentrated solutions for the first time.The sequence of amino acids was identified by liquid chromatography-ESI-mass spectrometry/mass spectrometry(LC-QTOF-MS/MS) and synthetic standards.The ACE inhibitory activity assay was determined for the peptide by using the modified HPLC method.The inhibition pattern on ACE and IC50 of the bioactive peptide were also studied in the paper.The results showed that the novel ACE inhibitory peptide,which was composed of3 amino acids,was discovered for the first time in Chinese Baijiu and the sequence of the peptide was Pro-His-Pro(PHP,MW;349 Da) with an IC50 value of 446 mM.In addition,Lineweaver-Burk plots suggested that PHP played the role as a noncompetitive inhibitor against ACE.
