蚕蛹蛋白免疫活性肽酶水解条件的优化

【摘要】 蚕蛹是一种优质的动物蛋白来源,但食用蚕蛹会引起部分人群过敏,存在食物安全问题,从而使蚕蛹在食品领域的应用受到限制。酶解可将蚕蛹蛋白分解成肽段,既能降低过敏的风险,又能获得具有功能活性的肽。本文选用菠萝蛋白酶、碱性蛋白酶、中性蛋白酶、风味蛋白酶和木瓜蛋白酶这五种酶对脱脂蚕蛹蛋白进行酶解。通过对小鼠脾细胞的增殖检测,选取最佳蛋白酶。在此基础上对最佳蛋白酶的四个单因素(酶和底物量,初始pH值,水解温度和底物浓度)进行优化,从而确定最佳的酶解条件。实验结果发现五种蛋白酶中碱性蛋白酶对脱脂蚕蛹蛋白的酶解效果最好。对碱性蛋白酶进一步优化,通过Box-Behnken实验和回归分析获得碱性蛋白酶酶解脱脂蚕蛹蛋白的最佳条件为酶解时间=1.96h,pH=8.03,水/底物=20.16。
【Abstract】 The silkworm pupa is a high-quality source of animal protein.However,it could cause the allergic reaction as a problem of food safety,so it can not be applied widespread in food industry.For the preparation of immunoactive peptide,we selected the five kinds of enzymes(bromelain,alkaline protease,neutral protease,flavourzyme,papain) to hydrolyze the defatted silkworm pupa protein.By the stimulation of spleen cell proliferation,the optimal enzyme were determined.On the basis of that,the optimal condition was identified with four singe factor of hydrolysis(E0/S0 PH,temperature and substrate concentration).The results showed that alkaline protease was the optimal enzyme and the optimum conditions of defatted silkworm pupa protein by alkaline protease enzyme was as that of enzymatic hydrolysis time = 1.96 h,pH = 8.03,water / substrate = 20.16.
