施用烟秆生物黑炭对烟田根际土壤微生物的影响

【摘要】 为研究烟秆生物黑炭对烟田根际土壤微生物的影响,采用田间随机区组小区试验,在起垄前对烟田土壤施用不同量的烟秆生物黑炭(0、10、40、80 t/hm2),在烟株旺长期采集根际土壤,采用高通量测序法测定根际土壤微生物。结果显示,施用生物黑炭后,根际土壤的微生物种类(OTU数)提高了26.4%。优势菌种中,变形菌门所占比例最大,达到47.19%~54.32%。施用生物黑炭增加了变形菌门、酸杆菌门、疣微菌门、拟杆菌门的相对丰度,减少了芽单胞菌门、绿弯菌门、硝化螺旋菌门、浮霉菌门的相对丰度。对于细菌类群研究还发现,在生物黑炭施用下,部分促生菌呈增长趋势。根际土壤养分检测结果显示,有机质、全氮呈增长趋势。综上所述,生物黑炭的施用影响了土壤的肥力,使土壤微生物的种类增加,并出现了有利植物生长的促生菌丰度的增加。
【Abstract】 The tobacco stalk-derived biochar was applied to the tobacco filed at levels of 0, 10, 40, and 80t/hm2, respectively. They were preceded in randomized block design before ridging. The effects of the biochar on rhizosphere microbes were investigated by the collection of the rhizosphere soils at vigorous growth stage and further test using high throughput sequencing. The results indicated that the amounts of the rhizosphere soil microbes increased after the applications of biochar, and the greatest increase(3.25 times) was achieved by the group at the level of 80 t/hm2. The operational taxonomic unit(OTU)increased by 26.4%. The OTU analysis on bacteria at 97% similarity demonstrated that there was no difference at the top 10 dominant phyla between the groups at different levels. The highest distribution was observed in Proteobacteria that accounted for 47.19% to 54.32% of the total bacteria. The increase of Proteobacteria, Acidobacteria, Verrucomicrobia, and Bacteroidetes and decrease of Gemmatimonadetes, Chloroflexi, Nitrospira, and Planctomycetes were positively associated with the application of biochar. This association was also applied to the increase of plant growth-promoting microorganisms(PGPMs). In addition, the soil organic matter, total nitrogen and available potassium were observed to increase significantly after the application by using the nutrition test of the rhizosphere soil.In conclusion, the soil fertility, the amount and diversity of soil microbe, and PGPMs were totally influenced by the application of tobacco stalk-derived biochar.
