抗痛风中药的体外活性筛选
Screening the anti-gout traditional herbs from TCM using an in vitro method

【摘要】 目的:评价33味中药提取物的抗炎作用和抑制黄嘌呤氧化酶(XOD)的活性。方法:通过尿酸钠(MSU)刺激人脐静脉血管内皮细胞(HUVEC)建立体外急性痛风模型,并测定细胞上清液和细胞裂解液中ICAM-1和IL-1β的表达水平来评价体外痛风模型的可行性;通过体外细胞模型筛选中药提取物及标准品对MSU刺激的体外急性痛风模型的抗炎作用:通过超高效液相色谱-质谱/质谱(UPLC-MS/MS)的方法监测黄嘌呤氧化生成尿酸的含量,反应中药提取物及标准品对XOD的抑制作用。结果:丹参、大黄、虎杖、卷柏、赤芍,尤其是银杏叶能够更好的抑制MSU诱导的炎症和抑制XOD活性,这与其含有的主要活性成分黄酮类和蒽醌类相关。
【Abstract】 The aim of this work was to evaluate the ability of 33 herbal extracts in inhibiting the acute inflammation and XOD activity.The anti-inflammation effects of the herbal extracts were detected by an in vitro cell model,which was established by stimulating HUVEC using MSU.In this model,the ICAM-1 and IL-1β were expressed,and the anti-inflammation effects of herbal extracts were evaluated by detecting the content changes of ICAM-1 and IL-1β in cell lysates and cell culture supemates using ELISA.Moreover,the UPLC-MS/MS method was used for the screening of XOD inhibitors in this research.The amount of uric acid from each analyte was directly detected using the multiple reaction monitoring mode.Results indicated that the anti-inflammation and enzyme inhibitory activities of the herbal extracts may be correlated with their bioactive components.The compounds of flavonoid and anthraquinone seem to be more effective on inflammation and inhibiting XOD.
