淀粉酶响应性Bola型超两亲分子

【摘要】 利用β-环糊精和一种Bola型共价两亲分子之间的主客体相互作用,我们制备了一种具有淀粉酶响应性的Bola型超两亲分子。Bola型共价两亲分子(BA)本身在溶液中可组装成片状聚集体,由于发色团的聚集淬灭效应,此聚集体呈现微弱的荧光。β-环糊精的加入可以使体系中形成Bola型超两亲分子,抑制BA分子的聚集,从而大幅提高体系的荧光强度。向体系中再加入α-淀粉酶后,随着β-环糊精持续被酶催化水解,超两亲分子逐渐解体,BA分子逐渐趋向于初始的聚集状态,体系的荧光强度也随之逐渐衰减。研究表明,此体系中荧光衰减的初始速率与α-淀粉酶的总活度呈现良好的线性关系,有望提供一种简便检测α-淀粉酶活性的新方法。

【Abstract】 An amylase-responsive bolaform supra-amphiphile was constructed from β-cyclodextrin and a bolaform covalent amphiphile based on host-guest complexation. The bolaform covalent amphiphile could self-assemble in solution, forming sheet-like aggregates and displaying weak fluorescence because of aggregation-induced quenching. The addition of β-cyclodextrin induced the formation of the bolaform supra-amphiphile, prohibiting the aggregation of the bolaform covalent amphiphile and leading to significant recovery of fluorescence. When α-amylase was added to degrade β-cyclodextrin, the fluorescence of the supra-amphiphile would be quenched gradually and significantly, and interestingly, the quenching rate linearly correlated to the concentration of α-amylase. This study enriches the field of supra-amphiphiles and may provide a facile way to estimate the activity of α-amylase.
