尿液中短链脂肪酸的高效萃取及测定

【摘要】 生物样品中短链脂肪酸(SCFAs)的含量与人的某些疾病如孤独症和肥胖紧密相关,但生物样本如尿液的基质复杂且SCFAs极易挥发导致前处理操作困难。现有的固相萃取前处理技术存在试剂消耗量大、操作繁琐而固相微萃取(SPME)技术存在重现性差、成本较高等问题。本文提出基于纳米纤维的固相萃取技术(PFSPE),制备的萃取柱仅需5mg吸附剂,微升量的洗脱剂,且操作简单、快速,易于批量操作。对比实验发现,聚苯乙烯(PS)纳米纤维对于多种SCFAs的萃取性能优于SPME。相比SPME,新方法对大多数SCFAs的检测信号能提高数倍,且方法的精密度也优于SPME。

【Abstract】 The content of SCFAs has an important relationship to human diseases such as autism and obesity,while the bio-sample like urinecontains very complex matrix,and the volatile of SCFAs makesthe pretreament of the sample extremely difficult.The tedious operation and poor reproducibility were exist in solid-phase extractionand solid-phase microextraction(SPME),respectively.In this work,a new method called packed fiber solid-phase extraction(PFSPE) was developed.The sorbent only needs 5 mg and the eluent is in microliter grade.Also the new method is simplicity of operation.The comparative experiments showed that the PFSPE method was better than SPME in adsorption quantity and precision.
