二维液相色谱-质谱联用对腔隙性脑梗死患者血浆的脂质组学分析

【摘要】 卒中是世界上致死和致残的一个主要原因[1]。探讨该疾病的早期诊断生物标志物及发病机制,对其诊断和治疗至关重要。本研究通过采用二维液相色谱-四级杆飞行时间质谱联用的方法分析了正常人和腔隙性脑梗死患者血浆中的脂质类化合物。经过初期的筛选及进一步的验证,我们找到了13个在两组间存在显著性差异的脂质分子。此后,通过二元逻辑回归分析,我们建立了一个包含葡萄糖基神经酰胺(38:2)、磷脂酰乙醇胺(35:2)、脂肪酸(16:1)和甘油三酯(56:5)这四种脂质化合物的"血浆生物标志物模型"。最后,基于这些实验结果,我们给出了在该疾病中可能受到扰动的脂质代谢通路。该研究表明了二维液相色谱-质谱联用的脂质组学分析方法在研究用于疾病诊断的生物标志物中的潜在应用价值,并为小血管性卒中早期诊断生物标志物及其发病机制的研究提供了更多的依据。
【Abstract】 Stroke is a major cause of mortality and long-term disability worldwide[1].The study of biomarkers and pathogenesis is vital for early diagnosis and treatment of this disease.In the present study,a continuous-flow two dimensional liquid chromatography- quadrupole time-of-flight mass spectrometry(2D LC-QToF/MS) method was employed to measure lipid species in human plasma,including healthy controls and lacunar infarction(LI) patients.As a result,13 lipid species were demonstrated with significant difference between the two groups after initial discovery phase and further validation phase,and a "plasma biomarker model" including glucosylceramide(38:2),phosphatidylethanolamine(35:2),free fatty acid(16:1) and triacylglycerol(56:5) was established.At last,related pathways of underlying pathophysiology were generated based on the lipidomic findings.This study demonstrates the promising potential of 2D LC-QToF/MS-based lipidomics approach in finding biomarkers for disease diagnosis and providing special insights into the metabolism of stroke induced by small vessel disease.
