三液相浮选-制备型高效液相色谱分离甘草中四种生物活性成分

【摘要】 本文建立三液相浮选-制备型高效液相色谱的方法分离纯化甘草中四种生物活性物质。在样品前处理部分,目标物采用三液相浮选的方法进行分离和富集,用10 ml乙酸乙酯作为上相,10 ml PEG1000的水溶液(w:w=1:1)作为中间相,调节溶液pH、盐浓度、氮气流速等参数,使甘草苷、芹糖甘草苷、甘草酸富集在PEG相中,乌拉尔甘草皂苷乙富集在乙酸乙酯相中。再采用制备型高效液相色谱,从浮选产物中依次分离纯化出甘草苷、芹糖甘草苷、甘草酸和乌拉尔甘草皂苷乙。三液相浮选-制备型高效液相色谱法可以高效分离甘草中四种生物活性物质,对于实现天然产物的资源化利用具有重要意义。
【Abstract】 Using three-liquid-phase flotation(TLPF),a new pretreatment method for separating and concentrating four bioactive compounds from the aqueous extract of Glycyrrhiza uralensis Fisch was developed,ethyl acetate was used as upper phase and PEG1000 was used as middle phase.By adjusting the pH,concentration of inorganic salt,nitrogen flow rate and flotation time,liquiritin,liquiritin apioside and glycyrrhizic acid were concentrated in upper phase and uralsaponin B was concentrated in middle phase respectively.After TLPF concentration,the flotation products were further purified by prep-HPLC.The research results indicated that the combination of TLPF and prep-HPLC is feasible and efficient for separation and purification of bioactive compounds from natural products.
