基于阳离子型水溶性聚噻吩衍生物的细胞中溶酶体ATP成像研究

【摘要】 在外在刺激下,星型胶质细胞和小神经胶质细胞的溶酶体能够通过钙离子调节的胞吐作用释放信号分子ATP,其它细胞(例如,He La细胞)是否也具有释放ATP的功能有待进一步研究。目前,还没有荧光探针应用于溶酶体ATP检测。我们合成了一种具有溶酶体靶向阳离子型水溶性聚噻吩衍生物(PEMTEI),并将其作为荧光探针应用于溶酶体ATP检测。PEMTEI对ATP具有高选择性和灵敏度,检测限为10-11 M。PEMTEI毒性低,生物相容性好,在细胞中的光学稳定性高,可实时监测溶酶体中ATP浓度的变化。此外,在药物刺激下He La细胞中的溶酶体能够通过钙离子调节的胞吐作用释放ATP。

【Abstract】 Lysosomes in astrocytes and microglia can release ATP as the signaling molecule for the cells through Ca2+-dependent exocytosis in response to various stimuli. However, the possibility that ATP releasing through lysosome exocytosis may be a general function in other cells(e.g., He La cells) has not been determined. At present, fluorescent probes that can detect ATP in lysosomes have not been reported. We have developed a water-soluble cationic polythiophene derivative(PEMTEI) that can be specifically localized in lysosomes and can be used as a fluorescent probe to sense ATP in cells. PEMTEI exhibits high selectivity and sensitivity to ATP and the detection limit of ATP is 10-11 M. The probe has been applied successfully to real-time monitoring of the change of ATP levels in lysosomes through fluorescence microscopy. We also demonstrated that lysosomes in He La cells can release ATP through Ca2+-dependent exocytosis in response to drug stimuli.
