饲草型小黑麦遗传图谱的构建及抗锈病基因的QTL定位

【摘要】 以性状差异较大的2个小黑麦品系为亲本进行杂交获得包含184个株系的F2分离群体为材料,利用ISSR分子标记筛选得到的14对多态性引物对小黑麦F2代群体进行扩增,运用Joinmap4.0作图软件对小黑麦F2代群体进行遗传连锁分析,构建了1张小黑麦遗传连锁图谱,并对抗锈病基因进行QTL定位分析,结果如下:该图谱包含7个连锁群(分别命名为LG1~LG7),连锁图谱总共有92个位点,标记间平均距离为5.90 cM,图谱总长度为542.9 cM;小黑麦F2群体184个单株锈病抗性的田间表型鉴定结果表明,锈病抗性在F2群体中呈连续变异,分布频率大致接近正态分布,可用于QTLs定位;运用Map QTL6.0软件对小黑麦F2群体的抗锈病基因进行QTL定位分析,共检测到6个QTLs,贡献率在5.1%~11.2%,分布在5个连锁群上,平均每个连锁群1.2个,QTLs在5个连锁群上的分布不均匀,连锁群LG5上分布最多,有2个QTLs。
【Abstract】 2 new triticale lines with significant differences in properties were used as parents for hybridization,these 184 plants in the F2 population as the materials in this experiment,Fourteen primers screened by the ISSR molecular marker were used to amplify the F2 generation,the genetic linkage analysis was carried out and 1 genetic linkage map was constructed using Joinmap4.0 software,and locate the QTLs related to the rust resistance in triticale,the results as follows:This linage map included 7 linkage groups(named LG1 ~ LG7),there were 92 loci,the averaged distance among markers was 5.90 c M,and the total length of map was 542.9 c M;Phenotype identification showed that characters related to the rust resistance in 184 plants of F2 population changed continuously and the distribution frenquency closed to the normaldistribution.Therefor,it was a suitable population to locate the QTLs related to the rust resistance in triticale;QTL results showed that there were 6 QTLs related to the rust resistance in triticale,contribution rate was 5.1% ~ 11.2%,these QTLs distributed on 5 linkage groups and there were 1.2 QTLs averagely on each linkage group,QTLs did not distribute evenly on 5 linkage groups,there were 2 QTLs distribute on LG5.
