正交设计优化常山总碱的超声提取工艺

【摘要】 目的:确定常山总碱超声提取的最佳工艺条件。方法:采用稀盐酸溶媒、超声提取法从中药常山中分离常山总碱,应用正交试验法确定其最佳提取工艺条件,采用分光光度法测定常山碱含量。结果:常山总碱最佳提取工艺为:提取温度50℃,料液比1:5,提取时间1 h;上述条件下常山总碱的提取率为2.57%,其中常山碱的含量为0.30%。结论:超声提取法用于常山总碱的分离,具有省时、节能、提取率高等优点。
【Abstract】 Objective To optimize ultrasonic extraction technology of total dichroa alkaloid(TDA).Methods:TDA of Dichroa febrifuga Lour were separated by dilute hydrochloric acid and ultrasonic extraction method.Its extraction process were optimized by orthogonal design.Dichroine content were determined by spectrophotometry.Results:The optimization process of TDA as follows:temperature 50℃,solid-liquid ratio 1:5,time 1 h.Under above conditions,TDA yield were up to2.57%) and the dichroine content were up to 0.30%.Conclusion:Isolation process of TDA is feasible by ultrasonic extraction method,which have advantages of time-saving,energy conservation and higher extraction rate.
