【摘要】 目的通过采用传统和快捷两种方法,建立小鼠急性心肌梗死模型,对两种方法制作的模型进行评估,比较两种方法的优缺点,为急性心肌梗死的深入研究寻找合适的小鼠模型制作方法。方法 40只C57/BI-6雄性小鼠随机分为两组:一组为传统方法组,另一组为快捷方法组,每组各20只。传统方法是将小鼠麻醉后进行口腔气管插管,结扎左冠脉前降支;快捷方法则无需气管插管,快速挤出心脏,结扎左冠状脉前降支。比较两种方法手术所用时间及模型的存活率,心电图评估模型成功与否,HE染色观察心肌组织病理学改变。结果传统方法手术用时20±5min,快捷方法用时仅1.3±0.2min;传统方法组模型的存活率75%,快捷方法组为90%;两组均出现ST-T抬高;HE染色两组心肌组织均出现变性坏死、空泡样变等病理改变。结论两种方法都能成功制备小鼠急性心肌梗死模型,传统方法用时长,存活率相对较低,但技术要求低,适合大多数研究者进行造模。快捷方法高效简洁,模型存活率高,特别适合大样本研究,但对研究者的手术技术要求很高,需要很高的娴熟度。更多还原

【Abstract】 Objective By using traditional and quick method,to establish acute myocardial infarction model in mice,to evaluate the both mice model,we can look for the right mice model method For the further study of acute myocardial infarction.Methods 40 C57/BL-6 male mice were randomly divided into two groups:a group for traditional methods,the other groups for quick methods,each group of 20.In the traditional method:after anesthesia,the mice were used oral endotracheal intubation,ligature left coronary artery anterior descending.In quick method,without endotracheal intubation,extrude the heart quickly,ligation coronary arteries left anterior descending.We compare the surgery time and the survival rate,assess electrocardiogram,and observe the myocardial tissue pathology change by HE staining.Results traditional method took about 20±5 min,quick method took 1.3±0.2 min.The survival rate of traditional methods group was 75%,quick method group was 90%.Both electrocardiogram appeared STT up.Both groups HE showed up Cavitation and pathological changes.Conclusion Both methods can successful made acute myocardial infarction model in mice.Traditional method need more time and survival rate is lower,but low technical required,and is suitable for most researchers.Quick method uses very little time and survival rate is higher,is especially suitable for large sample research.But quick method needs the researchers with very high surgical technique and skilled degrees.更多还原
