两种2型糖尿病动物模型的制备与比较
Preparation and comparison of two animal models of type 2 diabetes mellitus

【摘要】 目的比较KM小鼠与中国地鼠两种2型糖尿病动物模型在体质量、糖代谢、脂代谢与胰岛素抵抗等方面的差异。方法 KM小鼠与中国地鼠各30只,分别分为对照组(10只)与模型组(20只)。对照组饲以普通饲料,模型组用高糖高脂饲料结合STZ诱导建立2型糖尿病动物模型。检测各组动物体重,糖耐量,血液生化值以及血清胰岛素等指标,实验结束后取胰腺进行病理观察。结果 KM小鼠与中国地鼠都成功建立了血糖呈中轻度升高的2型糖尿病动物模型,且各自的成模率为70%和60%。在实验周期内,KM小鼠与中国地鼠模型组体重增长速度均呈现先快后慢的趋势;与对照组相比,两种动物CHOI与GLU以及KM小鼠的LDL均显著升高,AUG增大,FINS上升,HOMA-IR明显下降,其中,中国地鼠对照组在整个糖耐量实验中血糖变化较平稳;两种动物模型组的胰腺组织均出现轻微病变。结论 KM小鼠与中国地鼠模型组体态变化不一,代谢异常。KM小鼠呈肥胖体态,灌服葡萄糖后,个体血糖值差异较大;中国地鼠体态匀称不肥胖,灌服葡萄糖后,个体血糖值差异小,可能与其自身体内调节机制有关。
【Abstract】 Objective To compare the differences of body weight,glucose metabolism,lipid metabolism and insulin resistance between KM mice and Chinese hamsters with type 2 diabetes mellitus.Methods KM mice and Chinese hamsters were divided into control groups(n=10)and model groups(n=20).The model groups was fed with high glucose and high fat diet combined with STZ to establish the animal model of type 2 diabetes mellitus.The body weight,glucose tolerance,blood biochemical value and serum insulin were measured in each group.After the experiment,the pancreas was taken for pathological observation.Results KM mice and Chinese hamsters had successfully established animal models of type 2 diabetes mellitus with mild increase in blood glucose,and their respective molding rates were 70%and 60%.During the experimental period,the weight gain rate of KM mice and Chinese hamster in model groups showed the trend of the first rapid growth and then slowly.The CHOL and GLU in KM mice and Chinese hamsters,LDL in KM mice were significantly higher than those of the control groups,AUG and FINS increased,HOMA-IR decreased significantly.Among which the changes of blood glucose in control groups of Chinese hamsters were more stable.The pancreatic tissue of KM mice and Chinese hamsters were mild lesions in model groups.Conclusion In model groups,KM mice and Chinese hamsters were different in body changes,metabolic abnormalities.KM mice were obese body,after glucose infusion,the individual blood glucose difference is different.Chinese hamsters were not obese and symmetry,after filling glucose,individual blood glucose difference is small,may be related to its own body regulation mechanism.
