刀鲚Foxl2基因和Cyp19基因的分子克隆、组织分布表达、胚胎发育表达以及对性腺发育的影响

【摘要】 FOXL2在脊椎动物中作为一个很罕见的卵巢分化基因,能够对卵巢芳香化酶(CYP19)编码基因CYP19a起到激活转录和上游调控的作用。在硬骨鱼类中,FOXL2基因和CYP19基因已在许多鱼类中克隆分离得到,它在刀鱼性腺发育方面的生理学功能还有待研究。本实验采用RACE方法,获得了刀鱼FOXL2基因和CYP19基因的全长cDNA序列,长度分别为1494bp和1654bp。RT-PCR结果分析表明,FOXL2基因和CYP19基因在刀鱼的性腺中都呈现出卵巢的表达量要高于精巢的性别二态性表达方式,CYP19在卵巢不同发育时期的表达量在整个过程中呈现递增趋势,FOXL2基因和CYP19基因在刀鱼胚胎发育早期即被检测到,并随着幼体发育其表达量升高。上述研究结果说明,FOXL2基因和CYP19基因在刀鱼的性腺发育中发挥重要生理作用。
【Abstract】 FOXL2 acts as a rare ovarian differentiation gene in vertebrates, and plays a role in activating transcription and upstream regulation of ovarian aromatase(CYP19) encoding gene CYP19 a. In teleost fish, the FOXL2 gene and CYP19 gene have been cloned and isolated in many fish species, but its physiological function in the gonadal development of Coilia nasus remains to be studied. In this study, the full-length cDNA sequences of the FOXL2 gene and CYP19 gene were obtained with RACE method. The length of the cDNA was 1494 bp and 1654 bp, respectively.The result of RT-PCR analysis showed that FOXL2 gene and CYP19 gene showed the ovarian expression in the gonads of Coilia nasus, which was higher than that in the testis, the expression of CYP19 in the ovary at different developmental stages showed an increasing trend in the whole process, FOXL2 gene and CYP19 gene were detected at the early stage of embryo development and increased with larval development. The above results indicate that FOXL2 and CYP19 play important roles in gonad development of Coilia nasus.
