响应面优化蓝藻溶藻菌CZBC1的发酵工艺

【摘要】 为了提高蓝藻溶藻菌CZBC1的芽孢数量,开展其发酵工艺优化研究。在摇瓶培养条件下,通过单因素实验,筛选适合菌株CZBC1生长的最适碳源、氮源。再在单因素试验的基础上,选择四个条件(碳源、氮源、pH、培养时间)作为因素,采用Central Composite Design(CCD)试验设计确定关键因子的最佳数量水平,并用Desig-Expert 8软件进行回归分析,通过响应面分析获得菌株CZBC1的发酵工艺理论参数。研究发现,菌株CZBC1的最佳碳源、氮源分别为麸皮和糖蜜(1:1)、酵母膏。选择麸皮和糖蜜、酵母膏、pH、培养时间作为四因素,以菌株CZBC1的芽孢数为响应值,进行CCD试验设计,通过响应面分析得到发酵最佳的培养基。其组成为:麸皮和糖蜜6.84 g/L(mL/L),酵母膏3.36 g/L,pH 6.50,培养时间42.13 h,预测的芽孢数量为5.90×10~8 cfu/mL,显著高于未优化前(1.99×107 cfu/mL,P<0.01)。本研究可为蓝藻溶藻菌CZBC1的高效发酵培养提供有益的参考,也为进一步推动蓝藻溶藻菌制剂产品的制备与应用提供良好的基础。

【Abstract】 In order to improve the yield of the spores of algae-lysing bacteria CZBC1, response surface design was used to optimize its fermentation process. The single-factor experiment was initially used to find the optimal carbon source and nitrogen source. Then, using the Central Composite Design, the correlation equation between the most significant factors and the yield of the spores of algae-lysing bacteria was established by Response Surface Methodology. The results showed that the best carbon source and nitrogen source of Strain CZBC1 were bran and molasses(1:1), yeast extract, respectively. Choosing bran and molasses, yeast extract, pH and incubation time as four factors, the spore count of Strain CZBC1 as response value, the best fermentation medium was obtained by response surface analysis. Its composition included the bran and molasses 6.84 g/L(m L/L), yeast extract 3.36 g/L, pH 6.50, and 42.13 h of incubation time, and the number of spores of 5.90×10~8 cfu/mL, which was significantly higher than that before optimization.
