温度、流速对恩诺沙星在虹鳟体内药物代谢动力学的影响

【摘要】 众多环境因素会对药物在动物机体内的代谢产生不同的影响,为探明温度及流速对恩诺沙星在虹鳟体内药物代谢的影响,以虹鳟(Oncorhynchus mykiss)为试验材料,探究温度(5℃、10℃、15℃)和流速(8cm/s、16cm/s、24cm/s)对恩诺沙星在虹鳟体内吸收分布的影响。结果表明:在5℃,10℃和15℃水温下,恩诺沙星在虹鳟血浆中的达峰时间Tmax分别为8.67h、4.78h和2.39h;分布半衰期T1/2α分别为0.86h、0.08h和0.77h。肌肉和肝脏中药物代谢规律类似于血浆。实验温度下,表现为温度升高会加快药物的吸收和分布,即吸收分布速率增大,达峰时间缩短;。在8cm/s、16cm/s和24m/s三个流速下,恩诺沙星在虹鳟血浆中的达峰时间Tmax分别为8.57h、6.03h和4.04h;分布半衰期T1/2α分别为5.47h、2.16h和0.27h。水流速度加大,会导致药物吸收分布速率加快。本实验表明,在一定范围内温度的升高及水流速度的增加会加速恩诺沙星在虹鳟体内的吸收与分布。

【Abstract】 The pharmacokinetics of drug in animal was influenced by different environmental factor, in order to explore the effect of environment on pharmacokinetics of enrofloxacin in rainbow, this study explored the effect of temperature and light intensity on pharmacokinetics of enrofloxacin in rainbow. Results showed that at temperatures of 5℃,10℃ and 15℃,in plasma, the time-point of maximal concentration(Tmax) was 8.67 h, 4.78 h and 2.39 h, the distribution half-life(T1/2α) was 0.86 h, 0.08 h and 0.77 h, respectively. In muscle and liver, metabolism of drugs was similar to in plasma. Metabolism of enrofloxacin in rainbow showed: in a certain temperature range, the rate of drug absorption and distribution at higher temperature is larger than this at lower temperature; the time to peak was shorter at higher temperature. At water velocity of 8cm/s,16cm/s and 24cm/s, in plasma, the time-point of maximal concentration(Tmax) was 8.57 h, 6.03 h and 4.04 h, the distribution half-life(T1/2α) was 5.47 h, 2.16 h and 0.27 h. The rate of drug absorption and distribution at higher water velocity is larger than this at lower water velocity; the time to peak was shorter at higher water velocity.
