LC-MS/MS测定人血浆中紫杉醇、多西紫杉醇、长春花碱和异长春花碱
RAPID AND SENSITIVE LC-MS/MS METHOD FOR THE ANALYSIS OF PACLITAXEL,DOCETAXEL,VINBLASTINE AND VINORELBINE IN HUMAN PLASMA

【摘要】 目的:建立一种简单、快速和灵敏的LC-MS/MS方法测定人血浆中紫杉醇、多西紫杉醇、长春花碱和异长春花碱;方法:血浆样品经液液萃取后,流动相为乙腈-10mM醋酸铵(内含0.1%甲酸)(70:30),流速0.25mL/min,整个色谱方法运行4.5min,在Agilent G6410A串联质谱用正离子检测模式;结果:紫杉醇、多西紫杉醇、长春花碱和异长春花碱的线性范围分别为25.0-2500.0,10.0-1000.0,10.0-1000.0 and 10.0-1000.0 ng,mL;紫杉醇、多西紫杉醇、长春花碱和异长春花碱的r>0.990,日内和日间精密度均<9.2%;结论:该方法成功的用于非小细胞癌症患者服用紫杉醇、多西紫杉醇、长春花碱和异长春花碱的临床TDM检测。
【Abstract】 Objective:A rapid and sensitive analytical method using liquid chromatography coupled with electrospray ionization tandem mass spectrometry(LC-MS/MS) was developed for the determination of paclitaxel,docetaxel,vinblastine and vinorelbine in human plasma.Methods:A simple liquid-liquid extraction procedure was applied on only 100 μL plasma.Chromatographic separation of these anti-cancer drugs were achieved with an isocratic mobile phase consisting of acetonitrile/aqueous buffer(10 mM ammonium acetate and 0.1%formic acid)(70:30,v/v) at a flow rate of 0.25 mL/min in a short time(4.5 min).Results:The standard calibration curve for paclitaxel,docetaxel,vinblastine and vinorelbine in spiked human plasma were at about the ranges25.0-2500.0,10.0-1000.0,10.0-1000.0 and 10.0-1000.0 ng/mL respectively.The squares of the linear correlation coefficients were all over 0.99.The intra-day and inter-day relative standard deviation across three validations runs over the entire concentration range were less than 9.2%.Conclusions:The established method should be helpful for the pharmacokinetic monitoring of paclitaxel,docetaxel,vinblastine and vinorelbine in the human plasma of non-small-cell lung cancer patients.
