sCD83 modulates the immunological synapse formation between dendritic cells and T cells in autoimmune uveitis

【摘要】 Soluble CD83(sCD83), a soluble form of transmembrane CD83 molecule, has interesting therapeutic and immunosuppressive properties in the autoimmune disease, and transplant immunology. However, the molecular mechanism of sCD83 suppressing immune response is still unclear. Here, we show that DCs expressed high level of CD83 are upregu-lated in an experimental autoimmune uveitis model, which mimics the inflammatory status of autoimmune uveitis. Furthermore, in this study sCD83 ameliorated the symptoms of uveitis, and drastically reduced the number of infiltrated DCs and T cells in the infected eyes. Mechanistically, we show that sCD83 decreased the maturation of DCs and decreased the percentage of immunological synapse formation between DCs and T cells, by impairing the F-actin arrangement in the synapse resulting in the loss of ICAM-1 accumulation in the synapse. That is essential for the protective effects of sCD83 in experimental autoimmune uveitis.
