【摘要】 涎腺腺样囊性癌(SACC)是一种恶性肿瘤,周围神经侵袭是其显著特点。p53是一个重要的肿瘤抑制基因,p53基因突变在肿瘤发生和发展中发挥重要作用(胰腺癌、前列腺癌、头颈部癌症)。然而,p53基因在SACC嗜神经侵袭过程中的调控作用仍然是未知的。在目前的研究中,我们采用RNA干扰技术,抑制ACC-83细胞p53基因表达,研究p53在SACC嗜神经侵袭过程中所扮演的角色。我们的结果表明,p53基因表达下调可以显著诱导SACC-83细胞具备癌干细胞特性。同时,p53的下调也引起ACC-83细胞抗凋亡能力的提高以及体外转移及嗜神经侵袭能力的提高。因此,我们的研究表明,p53基因表达降低能够促进SACC-83细胞具备癌干细胞特性。本研究揭示了p53基因在SACC嗜神经侵袭过程中的重要调控作用,因此我们认为p53基因可能是一个新的临床治疗目标。更多还原

【Abstract】 Salivary adenoid cystic carcinoma(SACC) is a malignant tumor that is characterized by the perineural invasion(PNI).p53 is an essential tumor suppressor gene and the p53 mutations play a critical role in the tumor occurrence and progression(e.g.,pancreatic cancer,prostate cancer,and head and neck cancer).However,the regulatory role of p53 gene in SACC and the PNI process remain unknown yet.In the present study,we employed RNA interference technique to down-regulate the p53 gene expression in SACC-83 cells to explore the role of p53 in the PNI process.Our results showed that the down-regulation of p53 gene induced significant "cancer sternness" in SACC-83 cells.The down-regulation of p53 also caused lower apoptotic index of Annexin V-FITC/PI and lower number of second G0/G1 phase cell cycle in SACC-83 cells.Furthermore,the down-regulation of p53 gene resulted in significant increase of the PNI activity in SACC-83 cells.Thus,our findings suggest that down-regulation of p53 promoted in vitro PNI activity through "cancer sternness" in SACC-83 cells.The present study reveals the essential regulatory role of p53 in the PNI activity of SACC cells,and implies that p53 might be a new target gene for SACC clinical treatment.更多还原
