尼古丁在大鼠牙齿及牙槽骨分布的研究

【摘要】 目的:采用LC-MS/MS法测定大鼠牙齿及牙槽骨尼古丁含量,分析其在骨组织中的蓄积量,为探究尼古丁对大鼠骨组织的影响机制提供理论依据。方法:建立动物模型,将18只7周龄SD雄性大鼠分为空白对照组与3.2mg/kg高剂量组、0.8mg/kg低剂量组,每日进行尼古丁腹腔注射,3个月后处死,取大鼠上下颌切牙组织及牙槽骨组织,分别进行LC-MS/MS法测定,检测尼古丁在大鼠牙齿及牙槽骨中的蓄积情况。结果:将LC-MS/MS法检测数据进行参数统计分析,低剂量组尼古丁在大鼠牙齿及牙槽骨中蓄积量无统计学意义(P>0.05),高剂量组尼古丁可蓄积于大鼠牙槽骨中(P<0.05),在大鼠牙齿中的蓄积量无统计学意义(P>0.05)。结论:尼古丁在大鼠牙齿及牙槽骨内均能蓄积,其中在牙齿内的蓄积量与注射剂量无关,在牙槽骨内的蓄积量注射剂量呈正相关。发现尼古丁可蓄积在牙槽骨内,推测其参与了牙槽骨的平衡代谢。

【Abstract】 Objective:This research intends to establish the analysis method to determinate nicotine content in rat bone tissue through LC-MS/MS. In order to provide the critical basis to explore the effect mechanism and pathway of the nicotine in rat teeth and alveolar bone. Methods: Establish the analysis method to determinate nicotine content in rat bone tissue through LC-MS/MS. Eighteen health male SD rats of seven weeks of age were randomly divided into high dose group、low dose group and control group, the experimental group was tonicotine by intraperitoneal injection. Take tissues of rats’ teeth and alveolar bone. Detect of nicotine in the teeth and of rats alveolar bone by LC-MS/MS. Results: Establish a nicotine concentration in the tissue of the LC-MS/MS determination of analysis method. Take the data of LC-MS/MS method to parameter statistics analysis. Low dose group of nicotine in the rat teeth and alveolar bone volume no statistical difference(P>0.05), high dose group of nicotine can accumulate in alveolar bone of rats(P < 0.05), no statistical differences in volume in teeth(P>0.05). Conclusion:Nicotine can accumulate in the teeth and alveolar bone of rats, which in the teeth of volume has nothing to do with injection dosage, in alveolar bone volume dose were positively correlated. Found that nicotine can accumulate in the alveolar bone, speculated that its participation in the alveolar bone metabolism.
