外源油菜素内酯对根尖愈伤组织植株再生宽叶香蒲的重金属胁迫影响研究

【摘要】 通过根尖诱导宽叶香蒲愈伤组织,并选取含有不同配比的植物激素的分化培养基,建立宽叶香蒲高效愈伤组织诱导及苗分化再生体系。结果表明BA有利于根尖诱导宽叶香蒲愈伤组织的分化,IAA和NAA能够有效提高愈伤组织分化成苗的数量,KT则有利于分化苗的生长,吲哚乙酸能够促进5周龄宽叶香蒲苗生根和生长。对上述快速繁殖技术获取的大量宽叶香蒲苗进行重金属胁迫试验以及2,4-表油菜素内酯(2,4-EBR,简称EBR)对重金属胁迫的影响试验。结果表明:外源油菜素内酯存在的条件下,能够有效增强宽叶香蒲幼苗抗氧化系统(过氧化物酶、超氧化物歧化酶、过氧化氢酶等)的比活力,降低丙二醛含量,提高宽叶香蒲在重金属胁迫下的环境适应性能力,维持宽叶香蒲正常的生长代谢。
【Abstract】 A high-performance shoot regeneration and plantlet formation from root-culture-derived callus of Typha latifolia was established.Results showed that BA addition induced good differentiation of the calli and IAA and NAA additions could increase the number of shoots induced from calli,while KT benefited the shoots growth.5-week-old shoots were transferred to medium with indolbutyric,on which roots formation and growth were promoted.The rapid-propagation cattail seedlings were used to study the stress of heavy metals and the effect of EBR on alleciatin heavy metal stress.The resuts showed that the exogenous EBR could effectively improve the activities of anti-oxidizing enzyme systems(POD,SOD and CAT) and reduced the MDA contents in cattail seedlings.In conclusion,the exogenous EBR could alleviate growth of cattail seedlings under heavy metal stress.
