【摘要】 目的:探究冠心病患者与健康志愿者全基因组甲基化的差异。方法:取冠心病患者与健康志愿者血样提取DNA,用甲基化DNA免疫共沉淀测序的方法,比较冠心病患者与健康志愿者全基因组甲基化的差异。结果:甲基化峰扫描显示对照组的全基因甲基化水平低于冠心病组。在5’UTR,CDS,Intron三个基因功能元件上,对照组甲基化水平低于冠心病组。APQ1、SHB等基因启动子的甲基化在冠心病患者中显著降低;GRK5基因及chrX上多个基因的启动子甲基化在冠心病患者中显著增高。结论:冠心病患者与健康志愿者DNA甲基化水平有明显区别。
【Abstract】 Objective:To explore the genome-wide methylation differences between coronary heart disease patients and healthy volunteers. Methods:DNA was extracted from coronary heart disease patients and healthy volunteers’ blood, compare the genome-wide methylation differences between coronary heart disease patients and healthy volunteers with methylated DNA immunoprecipitation sequencing technology. Results:Methylation peak scans show whole genome methylation level in the control group is lower than the coronary heart disease group.In the 5’UTR,CDS,Intron functional elements, methylation level in the control group is lower than the coronary heart disease group. AQP1, SHB etc. gene promoter methylation level are significantly lower in patients with coronary heart disease. The gene promoter methylation level of GRK5 and several gene on chrX are significantly higher in patients with coronary heart disease. Conclusion:The DNA methylation level have obvious difference between coronary heart disease patients and healthy volunteers.
