蜈蚣珊瑚愈伤组织培养及丛生芽增殖体系优化
Optimization of Callus Culture and Shoot Multiplication System of Pedianthus tithymaloides ’Nana’

【摘要】 以蜈蚣珊瑚幼嫩叶片为外植体,采用正交试验设计,通过愈伤组织再分化和芽诱导直接增殖两条途径建立植株再生体系。研究表明:愈伤组织诱导的最佳培养基为MS+1.2mmg/L6-BA+0.5mmg/L2,4-D;不定芽增殖最佳培养基为MS+3.0mg/L6-BA+0.3mg/LNAA,最佳继代次数为4次;愈伤组织不定芽诱导最佳碳源为蔗糖;芽诱导的最佳培养基为MS+3.0mg/L6-BA+0.3mg/LNAA;生根培养最佳培养基为1/2MS+0.4mg/L IBA。本研究通过对蜈蚣珊瑚进行离体培养,初步获得组织培养各阶段最佳培养基配方,从而实现蜈蚣珊瑚离体快繁的目的,为蜈蚣珊瑚工厂化育苗提供理论基础及技术指导。
【Abstract】 In this study,we took leaves of Pedianthus tithymaloides cv.Nana as explants,The plant regeneration system was established by orthogonal experiment design and two ways of callus redifferentiation and bud induction direct multiplication.Results showed that the optimal medium of callus induction was MS + 1.2 mg/L6-BA +0.5 mg/L2,4-D.The best medium for adventitious bud proliferation was MS + 3.0 mg/L6-BA + 0.3 mg/L NAA,and the best subculture times were 4.the best carbon source for adventitious bud induced by callus was sucrose.The best medium for bud induction is MS +3.0 mg/L6-BA +0.3 mg/L NAA,The most suitable rooting medium was 1/2 MS +0.4 mg/LIBA.In this study,the explants of Pedianthus tithymaloides cv.Nana were cultured in vitro,and the optimum culture medium formula of each stage of tissue culture was obtained preliminarily,The purpose of this study was to achieve the rapid propagation,and to provide theoretical and technical guidance for the industrialization of plantlets of Pedianthus tithymaloides cv.Nana.
