南瓜组织DNA提取方法比较

【摘要】 南瓜(Cucurbita moschataDuch.)DNA提取是进行南瓜基因克隆、PCR扩增以及分子标记筛选的基础实验操作。本试验以南瓜杂交品种"百蜜一号"为试材,采用经典CTAB法、简化煮沸法和FTA法分别对南瓜根、茎、叶进行基因组DNA提取。结果表明:CTAB法适用于南瓜根、茎、叶的DNA提取,提取效果最好,但操作较为复杂;煮沸法操作过程也较复杂,从叶片提取的DNA质量相对较好;FTA法最方便、快捷,从叶片中提取的DNA质量相对较好。
【Abstract】 The DNA extraction is the basic experimental protocol of gene cloning,PCR reaction and molecular marker screening in Cucurbita moschata Duch.In this study,root,stem and leaf of pumpkin cultivar ’Baimi 1 hao’ were used to extract DNA through CTAB method,boiling method and FTA method.The results indicated that the DNA quality extracted from CTAB method is much better compared the other two.However,this method is complicated relatively.The DNA quality extracted from boiling method(except leaf) is not good enough and its process is complex.The DNA quality extracted from leaf through FAT method is acceptable and its process is relatively easy and efficient.
