梅花花香PmEGS1基因的克隆与序列分析

【摘要】 以‘三轮玉蝶’盛花期花朵为材料,根据梅花基因组数据库信息,通过RT-PCR方法克隆得到1个丁子香酚合成酶基因,命名为PmEGS1。该基因位于梅花的第2条染色体上,包含927bp的开放阅读框,编码308个氨基酸,编码蛋白质分子量约为33.9kDa,理论等电点为5.61,不稳定系数为29.01,属于酸性稳定的亲水性蛋白。DNAMAN比对和系统进化树构建结果表明,基因的cDNA及其推导的氨基酸序列均与草莓、月季、罗勒、仙女扇、矮牵牛等物种中的EGS具有高同源性,且在EGS基因所特有的结构域KQVDVVIS和KIIAAIK上高度保守。
【Abstract】 According to the P.mume genome database,a eugenol sythase gene named PmEGSl was cloned Irom Primus mume’ Sanlunyudie’ through RT-PCR method.The gene was located on the No.2 chromosome.It contained 927 bp ORF,encoding308 aa with a molecular mass of 33.9 kDa.The encoded protein was acidic stable hydrophilic with PI 5.61 and Ⅱ 29.01.Analysis of DNAMAN and phylogenetic tree showed that cDNA and the deduced amino acid sequence contained the characteristic conserved domains KQVDVVIS and KIIAAIK,and clustered with other EGS genes with high homology derived from Fragaria ananassa,Chinese rose(Rosa hybrida and Rosa chinensis),Ocimum basilicum,Clarkia breweri,Petunia hybrid and etc.
