洋桔梗叶片高频再生体系建立的研究

【摘要】 洋桔梗(Eustoma grandiflorum)花色丰富、姿态优美,是一种新型高档切花。本研究以洋桔梗品种‘优胜’(绿色)叶片为外植体,筛选最适灭菌条件,确定最适愈伤组织诱导培养基:MS+0.5mg·L-16-BA+0.1mg·L-1NAA,诱导率为83%;最适分化培养基:MS+0.5mg·L-16-BA+0.1mg·L-1IAA+1.0mg·L-1GA3,增殖系数为5.58;最适生根培养基:1/2MS+0.1mg·L-1NAA+1.0mg·L-1IBA,生根率为100%,平均每株根数为19.55。初步建立了洋桔梗再生体系,以期为洋桔梗遗传转化提供技术支撑。
【Abstract】 Lisianthus(Eustoma grandijlorum),with rich color,graceful,is a new type of cut flowers.This experiment used orthogonal experimental design,filtrating optimal mediums of differential for adventitious buds and roots,explants of E.grandijlorum were sterilized and callus were inducted to proceed genetic transformation and germplasm of Lisianthus.What results showed is that the optimal sterilization methods to establish leaf regeneration system of E.grandijlorum ’ superior’(green):75%alcohol with 2%NaClO disinfected for 20 min.MS contains 0.5mg/L 6-BA +0.1mg/L NAA with PH value of 6.3 is most suitable for inducing callus.Best medium for adventitious buds is MS +0.5mg · L-16-BA +0.1mg · L-1IAA + 1.0mg ·L-1 GA3.1/2MS+ 0.lmg- L-1NAA+ 1.0mg · L-1 IBA or 1/2MS + 0.1 NAA mg · L-1 +1.0 NAA mg · L-1 are best rooting,,E.grandijlorum regeneration system was established,with a view to laying the theoretical foundation for genetic transformation.
