转Bt基因水稻田间目的基因表达情况及抗螟虫性
Target gene expression and stem borer resistance of insect-resistant super japonica rice transformed with cry1C~* gene in field

【摘要】 为了研究转Bt基因水稻在田间目的綦因crylC*的表达及对螟虫的抗性,本研究以10份转CrylC*基因的抗虫水稻品系为材料,在水稻抽穗期,利用实时荧光定量PCR方法检测叶片、茎鞘和幼穗等器官目的基因mRNA,利用酶联免疫吸附(ELISA)法检测叶片、茎鞘和幼穗中及收获后糙米的CrylC蛋白,利用田间目测调查二化螟危害的白穗率。结果显示,转基因水稻抽穗期不同品系及不同器官crylC*基因旧间表达量明显不同,crylC*基因mRNA表达量叶片>茎鞘>幼穗,Bt蛋白质表达量叶片>茎鞘>幼穗>糙米。转基因水稻田间目的基因表达,抽穗期叶片、茎鞘和幼穗目的基因mRNA表达量与对应器官蛋白质表达量间存在极显著正相关关系,相关系数依次为r叶茎0.823、r叶穗0.878及r茎穗0.874;叶片、茎鞘和幼穗的Cry1C蛋白质含量和糙米Bt蛋白质含量也呈显著或极显著正相关,相关系数分别是r叶米0.878、r茎米0.646、r穗米0.653。在本研究范围内,不论转基因粳稻植株Cry1C蛋白质含量高或低的品系,田间均表现为高抗二化螟。
【Abstract】 In order to study the expression of crylC* and the resistance to stem borer in Bt transgenic rice,10 rice cultivars with crylC* gene were used as materials,and the results were detected by real-time fluorescence quantitative PCR The CrylC protein in leaves,stem and sheath and young spikelets and post-harvest brown rice were detected by enzyme-linked immunosorbent assay(ELISA).The results were analyzed by field visualization.rate.The results showed that the expression of crylC* gene in different lines and different organs of transgenic rice was significantly different,and the expression of crylC* gene mRNA> stem and sheath> young spike and Bt protein> leaf> stem and leaf> young spike> brown rice.There was a significant positive correlation between the mRNA expression level and the expression level of the corresponding organ protein in the transgenic rice field,and the correlation coefficient r was r叶茎0.823、r叶穗0.878 and r茎穗0.874 respectively.The CrylC protein content and the brown protein Bt protein The correlation coefficient r was r叶米0.878、r茎米0.646、r穗米 0.653 respectively.In the range of this study,regardless of the high or low protein content of CrylC protein in transgenic japonica rice,the field showed high resistance to Chilo suppressalis.
