REM-PCL对脊髓缺血再灌注损伤中脊髓微循环的影响

【摘要】 目的观察脊髓缺血再灌注损伤(SCIRI)中脊髓微循环血流量(SCMBF)、内皮素1(ET—1)及其ET—1相关蛋白的变化,探讨REM—PCL对SCIRI的影响及其意义,从而为临床防治SCIRI探索新的途径。方法 30只健康新西兰大白兔随机分为假手术组(S组,n=10)、对照组((组)和REM—PCL组(RP组,n=10)。采用肾下腹主动脉阻断法,建立脊髓缺血再灌注损伤模型。结果 C组NSE、ET—1、ET—1、ROS、ET—1、ECE和ETAR表达明显增加(P<0.01),SCMBF和MOR表达明显降低(P<0.01),脊髓灰质的病理损害严重(P<0.01)。RP组SCMBF和MOR表达明显增加(P<0.01),而ET—1、NSE、ET—1蛋白、ROS、ET—1、ECE和ETAR表达明显降低(P<0.01),脊髓灰质的病理损害程度明显轻于C组(P<0.01)。结论应用REM—PCL可以减轻ET—1的缩血管损伤等生物效应,对脊髓微循环具有保护作用,进一步证实ET—1相关蛋白和内源性MOR参与脊髓缺血再灌注损伤信号转导。
【Abstract】 Objective 30 healthy New Zealand white rabbits were randomly divided into sham operation group(S group,n= 10),control group(C group)and REM—PCL group(RP group,n=10).The model of spinal cord ischemia reperlusion injury was established by using the method of abdominal aortic occlusion.Result The level of MOR and SCMBF were significantly lower in C group during reperlusion than those in group RP(P<0.01).The level of NSE.ET—1.ROS.ET—1,ET—1,ECE and ETAR expression were significantly higher in C group during reperlusion than those in group RP(P<0.01).The pathological damage to the gray matter of spinal cord was more serious in C group during reperlusion than that in group RP(P<0.01).Conclusion REM—PCL can protect spinal cord microcirculation by alleviating the reduction of SCMBF and the raise of ET—1 during SCIR,which may have therapeutic benefits Tor ischemia/reperlusion—induced spinal cord injury to some extent.These ascertains endogenous ET—1 can cause spinal cord microcirculatory lesion through its effects of vasoconstricting and reducing SCMBF by ET—1 mediating vasoconstriction.These suggests ET—1 associated protein and endogenous MOR were involved in the signal transduction of SCIRI.
