普洱茶、绿茶水提物对非酒精性脂肪肝大鼠肠道脂肪吸收及粘膜屏障的干预研究

【摘要】 研究普洱茶、绿茶水提物调控非酒精性脂肪肝大鼠肠道长链脂肪酸吸收及其可能机制。采用SD大鼠48只,随机分成正常对照组、脂肪肝模型组、普洱茶干预组和绿茶干预组,每组12只。实验第8周末处死所有大鼠,测定大鼠体重、肝湿重及血清LDL-C水平,定量PCR测定大鼠肠道黏膜脂质吸收相关蛋白(CD36)、紧密连接相关蛋白(occludin、ZO-1)和肿瘤坏死因子α(TNF-α)基因表达水平。结果表明,与模型组相比,普洱茶组大鼠体重、肝湿重和血清LDL-C水平分别下降16.12%、15.35%和42.59%,绿茶组肝湿重和血清LDL-C水平分别下降21.98%和51.85%;普洱茶组、绿茶组大鼠小肠组织CD36、TNF-αmRNA表达水平明显降低,occludin、ZO-1mRNA表达水平明显升高;而且普洱茶干预组的CD36、TNF-αmRNA表达水平明显低于绿茶干预组,occludin、ZO-1mRNA表达水平两组间无明显差异。研究表明,普洱茶及绿茶对NAFLD大鼠肠道长链脂肪酸吸收及肠道紧密连接蛋白有一定程度的调控作用,尤其是普洱茶对NAFLD早期发生发展中游离脂肪酸过度沉积的"第一次打击"有防治价值。
【Abstract】 Objective:To study the possible interventional mechanism of pu’ erh tea and green tea water extract on long chain fatty acids absorption and metabolism and intestinal tight junction protein in non-alcoholic fatty liver disease(NAFLD) rats.Methods:48 SD rats were randomly divided into four groups,namely,normal group(NG),NAFLD group,Pu-erh tea group(PTG) and green tea group(GTG),12 rats each group,all rats were killed at 8 weeks,intestinal CD36,occludin,ZO-1,TNF-α mRNA were detected by RT-PCR Results:Compared with NAFLD group,the intestinal CD36,TNF- a mRNA expression level in PTG and GTG notably decreased(P<0.01),while the intestinal occludin and ZO-1 mRNA expression level significantly increased compared with NG(P<0.01).Compared with GTG,the expression level of CD36 and TNF- a increased remarkable,while the distinctions of occludin and ZO-1 were not significant.Conclusions:Pu-erh tea and green tea water extract can regulate gene expression levels of CD36,occludin,ZO-1,TNF-α to some extent,pu-erh tea was stronger than green tea on the effect of regulating the expression levels of CD36 and TNF-α,The study also suggested that tea,especially pu-er tea,may be effective in prophylaxis against "the first attack " in the early stage of NAFLD.
