鲤鱼汤对阿霉素肾病大鼠肾脏水通道蛋白表达的影响

【摘要】 目的:研究鲤鱼汤对阿霉素肾病大鼠肾脏水通道蛋白(AQP)表达的影响,探讨其治疗肾病水肿的可能机制。方法:成年健康Wistar大鼠50只,随机分为正常组、模型组、福辛普利组、鲤鱼汤高剂量组、鲤鱼汤低剂量组。经尾静脉注射阿霉素制备肾病综合征模型,应用鲤鱼汤干预治疗,生化法检测大鼠12h尿蛋白排泄量,血清白蛋白、总蛋白以及钾、钠、氯水平,HE染色观察肾脏组织病理变化,免疫组织化学方法检测肾脏组织中.AQP1-3表达水平。结果:①造模后模型组大鼠尿蛋白排泄量随时间延长逐渐升高,但鲤鱼汤治疗组未见显著升高(F=5.23~41.89,P<0.05)。②模型组大鼠血清白蛋白和总蛋白较对照组显著降低,鲤鱼汤治疗组较模型组均显著升高(F=13.12~15.48,P<0.05)。模型组大鼠血清钾、钠和氯水平较对照组显著升高,鲤鱼汤高剂量治疗组比模型组显著降低(F=3.42~3.96,P<0.05)。③模型组大鼠肾组织AQP1、AQP2、AQP3表达较对照组显著增强,鲤鱼汤治疗组较模型组显著减低(F=3.97~6.19,P<0.05)。结论:鲤鱼汤能通过下调肾脏AQP1、AQP2和AQP3表达.减少尿蛋白的排泄量,减少肾脏对水液的重吸收而缓解肾性水肿。

【Abstract】 Objective To observe the effects of Cyprinus carpio decoction on the expression of aquaporins in adriamycin-induced nephropathy rats and to explore the therapeutic mechanism on nephrotic edema.Methods Total of 50 Wistar rats were randomly divided into normal group,model group,fosinopril group,Cyprinus carpio decoction treated with high dose group and low dose group consisting of10 rats respectively.Nephropathy models were established by injecting adriamycin through tail vein and treated with Cyprinus carpio decoction.Urinary protein excretions in 12 h,serum albumin,total serum protein,serum sodium and potassium were measured by biochemical assay.The pathological changes and the expression of AQP1-3 in kidneys were respectively detected by HE stain and immunohistochemiscal assay.Results ① The urinary protein excretion in 12h(proteinuria) increased significantly along the time longed modeling,while no significant increasing in Cyprinus carpio decoction treated group(F=5.23-41.89,P<0.05).②The serum albumin and total protein in model group were significantly lower than that in normal group,but that in Cyprinus carpio decoction treated group were higher than that in model group(F=13.12-15.48,P<0.05).The serum sodium and potassium in model group were higher than those in normal group,while that in Cyprinus carpio decoction treated group were higher than that in model group(F=3.42-3.96,P<0.05).③ In model group,the expressions of AQP1-3 in the renal tubule and collecting duct cells increased significantly than those in normal group,and those in Cyprinus carpio decoction treated group decreased than those in model group(F=3.97-6.19,P<0.05).Conclusions Cyprinus carpio decoction could reduced the urinary protein excretion and alleviate pathological lesion and edema with adriamycin-induced nephropathy by decreasing the expressions of AQPs in kidneys.
