DS-1-47促着床的靶点分子机制筛选研究
Study on the Molecular Mechanism of DS147 in Promoting Embryo Implantation

【摘要】 目的:研究DS-1-47的促着床作用机制。方法:实验选取两种方法建立了小鼠不孕模型运用蛋白组学激光捕捉显微切割技术-二维电泳-质谱(laser capture microdissection-double dimension Electrophoresis-Mass spectum,LCM-DE-MS)技术路线研究分析DS-1-47作用前后在子宫组织细胞中受调节的蛋白分子,以筛选得到DS-1-47促进着床、调控着床的关键分子,从而为深入准确阐明其药效作用的分子调控机制和作用靶点。结果:CM组上调但是CZ组下调的蛋白质被鉴定为血液结合素,肌肽原-1、丝氨酸蛋白酶抑制剂A3K、α-1-抗胰蛋白酶1-3,结合珠蛋白、延伸因子-1-△、膜联蛋白A5、血清淀粉样蛋白P-成分;CM组下调但是CZ组上调的蛋白质为肝羧酸酯酶、丝氨酸蛋白酶抑制剂、载脂蛋白A-I。IM组上调但是IZ组下调的蛋白质为胶原α-1(Ⅵ)链、角蛋白,Ⅱ型骨架7、角蛋白,Ⅰ型骨架14、肌球蛋白调节轻链12B、肌球蛋白调节轻多肽9、热休克蛋白β-7、可能C->U-编辑酶APOBEC-2;IM组下调但是IZ组上调的蛋白质为载脂蛋白A-I,钙调节蛋白-3、增殖细胞核抗原、L-木酮糖还原酶、钙结合蛋白。结论:初步确定钙结合蛋白、载脂蛋白A-I和膜联蛋白A5是复方DS-1-47促进着床的可能重要靶点。
【Abstract】 Objective:To identify the mechanism for promoting the implantation of DS-l-47.Methods:This study chose two methods to establish the infertility model of mice.Using the technical route of proteomics laser capture micro microdissection-double dimension electrophoresis-mass spectrometr(LCM-DE-MS)to study and analysis the protein moleculars that were regulated by DS-1-47 in uterine tissue cells,and to screen the key molecules which promote implantation.So as to further clarify the molecular mechanism and function targets of the drug.Results:These proteins which were up-regulated in CM group but down-regulated in CZ group were identified as hemopexin,carnosine-l,serine protease inhibitor a3 k,α-l antitrypsin 1-3,haptoglobin,elongation factor-1-Δ,membrane associated protein A5,serum amyloid amyloid P-component.These proteins which were down-regulated in CM but up-regulated in the CZ group were identified as liver acid esterase,serine protease inhibitor,and apolipoprotein A-I.These proteins which were up-regulated in EM group but down-regulated in IZ group were collagen-1(Ⅵ)chain,keratin,Ⅱ type framework 7,keratin,Ⅰ type framework 14,myosin regulatory light chain 12 B,myosin light polypeptide 9,heat shock protein β-7,may be U-C-> editing enzyme APOBEC-2.These proteins which were down-regulated in IM group but up-regulated in IZ group were apolipoprotein A-I,Calcium regulated protein-3 proliferating cell nuclear antigen,L-Xylulose reductase,calcium binding protem.Conclusion:Preliminary determination that calcium binding protein,apolipoprotein A-I and annexin A5 are important targets for DS-1-47 to promote implantation.
