洁泽1号对阴道乳酸菌及免疫蛋白SP-D、sIgA及SLPI影响的实验研究

【摘要】 目的:研究中药复方洁泽1号对生理状态及白色念珠菌性阴道炎小鼠阴道产过氧化氢乳酸菌、pH值、过氧化氢浓度、SP-D、sIgA及SLPI的影响,评价洁泽1号对阴道内环境的影响。方法:1.将正常小鼠分为正常组、10%N-9组、空白凝胶组、2%洁泽1号组(2%JZ1)、1%洁泽1号组(1%JZ1),每组设24h和72h两个亚组,分别于给药后24h和72h观察阴道pH值、产过氧化氢的乳杆菌、过氧化氢浓度及SP-D影响;2.建立白色念珠菌性阴道炎小鼠模型,实验分为正常对照组、模型组、空白凝胶组、联苯苄唑组、2%洁泽1号组(2%JZ1)、1%洁泽1号组(1%JZ1)。分别于造模前24h和治疗的第0、4、8、12天观察小鼠阴道阴道pH值、产过氧化氢的乳杆菌,治疗的第0、12天检测小鼠阴道灌洗液中sIgA的表达,治疗第12天检测阴道组织中SLPI的表达。结果:生理状态下1.使用洁泽1号与正常组相比,小鼠阴道pH值、产过氧化氢乳酸菌、过氧化氢浓度均无明显变化,对阴道组织SP-D表达无影响;2.白色念珠菌性阴道炎病理状态下,模型鼠阴道乳杆菌数量减少,sIgA和SLPI表达升高,2%JZ1组治疗后各指标恢复到正常水平(P>0.05);1%JZ1组小鼠虽各个指标有改善,但未恢复到正常水平,(P<0.05)。结论:生理状态下,洁泽1号对阴道内环境无显著性影响,对白色念珠菌性阴道炎小鼠,有效治疗剂量的洁泽1号能增加阴道乳酸菌数量及上调相关免疫蛋白的表达,通过促进阴道菌群平衡,提高阴道局部固有免疫因子的表达可能是其治疗白色念珠菌性阴道炎的作用机制之一。
【Abstract】 Objective:Weinvestigated the effect of JieZeNO.l gel on vaginal hydrogen peroxide-producing lactobacillus,pH value,the concentration of hydrogen peroxide and vaginal surfactant protein D(SP-D),secretory immunoglobulin A(slgA) and secretory leukocyte protease inhibitor(SLPI) under the physiological state and pathological condition of WC to evaluate the effects of JieZeNO.l gel on the vaginal microenvironment.Methods:The normal mice were divided into groups of normal,10%N-9,blank gel,2%JZ1 and 1%JZ1,and each group was set for 24 h,72h two subgroups.The vaginal pH value,hydrogen peroxide-producing Lactobacillus,the concentration of hydrogen peroxide and expression of SP-D were observed at 24 h and 72 h after administration.The mouse model of WC was established,and the mice were divided into groups of normal,model,blank gel,bifonazole,2%JZ1 and 1%JZ1.The vaginal pH value and hydrogen peroxide-producing lactobacillus were observed 24 hours before treatment and the day 4,8,12 after treatment.Expression of sIgA protein in vaginal lavage was detected after 12 days of treatment.The expression of SLPI protein in the tissues of the vagina was detected.Results:There were no significant changes in 2%JZ1 and 1%JZ1 groups of vaginal pH value,hydrogen peroxide-producing lactobacillus,the concentration of hydrogen peroxide and expression of SP-D compared with those of the normal group under physiological state.Under the pathologic state of WC,vaginal pH value of model group increased,the number of hydrogen peroxide-producing lactobacillus for model group decreased,and the protein expression of slgA and SLPI of model group increased.After treatment,vaginal pH value,hydrogen peroxide-producing lactobacillus,the concentration of hydrogen peroxide and expression of SP-D and SLPI of 2%JZ1 group recovered to the normal level.Conclusion:Under physiological conditions,vaginal use of JieZeNO.l gel had no significant influence on the vaginal environment,and under the pathological condition of WC,promoting the vaginal flora balance and improving the expression of the local innate immune factor may be the part of the mechanism of JieZeNO.1 gel treatment of WC.
