去盆腔神经大鼠结肠动力变化及其黏膜TRPA1表达
A novel insight from a compensatory mechanism of colonic transit and expression of TRPA1 in a rat model of transection of pelvic nerves

【摘要】 目的:探讨去盆腔神经模型大鼠结肠动力恢复与结肠黏膜TRPA1表达变化的关系。方法:成年大鼠96只随机分为两部分:结肠动力检测部分42只,分为手术组与假手术组,利用放射性同位素法(51Cr)测定术后1、3、7天结肠传输功能;TRPA1检测部分54只,分为对照组、假手术组及手术组,利用Western Blotting检测术后1、3、7天近端及远端结肠黏膜TRPA1的表达情况。结果:与假手术组相比,手术组术后第1天结肠传输显著变慢[(4.91±0.41)vs(5.76±0.85),p<0.05],术后第3天结肠动力逐渐恢复[(5.58±0.36)vs(7.50±0.53),p<0.01],术后第7天进一步恢复,与假手术组无显著差异[(6.44±0.78)vs(7.31±0.46),p>0.05]。Western blot结果显示:手术组远端结肠黏膜TRPA1表达在术后第一天显著低于假手术组[(0.39±0.12)vs(0.68±0.11),p<0.01],术后第3天有所增加,仍低于对照组[(0.78±0.10)vs(0.98±0.12),p<0.01],术后第7天显著增加,与对照组差异无统计学意义[(1.06±0.13)vs(1.11±0.16),p>0.05]。结论:去盆腔神经支配后,大鼠结肠动力显著减弱,结肠黏膜TRPA1表达下调,均随时间推移逐渐恢复;这种结肠动力的适应性恢复与远端结肠黏膜TRPA1的表达变化有关。
【Abstract】 Objective To explore the relationship between recovery of colonic transit following transection of pelvic nerves in a rat model and the expression changes of transit receptor potential ankyrin 1(TRPA1) in the colon mucosa.Methods Ninety-six Sprague- Dawley rats divided into the sham operation group and the operation of transection of pelvic nerves group underwent placement of a tunneled catheter into the proximal colon.Colonic transit was assessed respectively on the postoperative 1,3,and 7 days by injecting and calculating the geometric center(GC) of the distribution of 51Cr after 3h of propagation.With the applications of Western blotting,expression of TRPA1 were determined in the colon mucosa.Results On the 1st postoperative day,transection of pelvic nerves significantly delayed colonic transit(GC=4.91 ± 0.41,p<0.05) compared with the sham operation(GC=5.76 ± 0.85).Unexpectedly,a significant trend toward recovery was recorded in the PN transection groups on the 7th day.On the1 st postoperative day,TRPA1 expression in the transection of pelvic nerves group were significantly lower in the proximal colon mucosa when compared with that in the sham operation[(0.39 ± 0.12)vs(0.68 ± 0.11),p<0.05)].On the 3st postoperative day,TRPA1 expression in the transection of pelvic nerves group were still significantly lower in the proximal colon mucosa when compared with that in the sham operation[(0.78 ± 0.10)vs(0.98 ± 0.12),p<0.05)].However,when the 7th postoperative day arriving,the TRPA1 expression in both groups represented a recovery trend and there was statistically no difference between them[(1.06 ± 0.13)vs(l.ll ± 0.16),p>0.05)].Conclusion Injury to pelvic nerves adversely affects colonic transit and expression of TRPA1 in mucosa.With a compensatory mechanism from the intestinal itself,these alterations in intestinal motility function mormalize over time suggesting expression of TRPA1 in mucosa plays a crucial role in the recovery of intestinal motility function.
