用于特异性检测溶酶体内硫醇的双光子荧光探针

【摘要】 体内硫醇作为多肽的组成成分在重大疾病的发病机制中起到重要作用[1]。此外,双光子荧光成像具有光毒性低,三维空间定位好,渗透能力强和自吸收能力弱等优点[2]。然而,这类检测硫醇的探针仍然是罕见的。在本工作中,我们设计合成了一例以萘酰亚胺为母体,连接2,4-二硝基苯磺酰氯和吗啉环的双光子荧光探针TC[3],对硫醇表现出高选择性和灵敏度;并且该探针具有优异的双光子性能和低细胞毒性,在双光子激发的荧光成像过程中,可以有效避免生物样品的背景荧光干扰。更重要的是,它可以定位于细胞的溶酶体,进而有利于实现了对该细胞器功能的阐释。
【Abstract】 Lysosomal thiols are components of many petides that play key roles in the pathogenesis of serious diseases[1].Moreover,two-photon microscopy offers advantages of less phototoxicity,better three dimensional spatial localization,deeper penetration depth and lower self-absorption[2].However,such fluorescent probes for thiols are still rare.In this work,an efficient two-photon fluorophore 1,8-naphthalimide-based probe TC[3] conjugating a2,4-dinitrobenzenesulfonyl chloride and morpholine was designed and synthesized,which exhibited high selectivity and sensitivity.Moreover,TC has outstanding two-photon performance and low cytotoxicity.It can avoid the background fluorescence interference of the biological samples effectively during the use of two-photon microscopy.Importantly,fluorescent probe TC that response to biothiols specifically in the lysosome is of interest to elucidate organelle functionality.
