钆/铕配合物-锂皂石磁共振/荧光双模态探针用于抗坏血酸检测

【摘要】 荧光/磁共振双模态技术受到了广泛关注。当前,基于稀土的荧光/磁共振探针主要用于生物成像,而用于检测的工作未见报道。我们通过离子交换方法将钆/铕配合物引入锂皂石片层内,制备了一种有机-无机杂化发光/磁共振材料(Gd-/Eu-DPA@Lap)。该材料具有良好的磁共振/荧光活性,对抗坏血酸(AA)具有特异性响应能力,检测限达到0.1?M(荧光模式)和0.2?M(磁共振模式),并实现了荧光/磁共振信号的交叉验证。此外,通过干扰实验发现:酸性和谷胱甘肽(GSH)等还原性物质对该材料的影响较小,而在盐酸和GSH共存的条件下出现与AA相类似的结果,说明该材料的检测机理源于酸性和还原性的共同作用。该杂化材料具有制备方法简便、毒性低、灵敏度高等优点,在生物传感领域具有重要的发展潜力。

【Abstract】 Magnetic resonance/Fluorescence(MR/FL) bimodal technology has ignited intense research interest worldwide. Recently, rare earth based MR/FL probes are used for biological imaging. However, the fabrication of MR/FL bimodal sensor is still challenging. Herein, a novel organic-inorganic hybrid material(Gd-/Eu-DPA@Lap) was built from gadolinium/europium complexes and laponite in ion-exchange method. The resulting material had good MR/FL activity and showed a highly selective to ascorbic acid(AA). This sensor showed a detection limit of 0.1 ?M in FL mode, while the MR mode exhibited a detection limit of 0.2 ?M. The cross-validation of MR/FL was also achieved to obtain detailed information. Furthermore, we found that hydrochloric acid(HCl) and glutathione(GSH) have little influence on hybrids, while it could achieve similar result to AA upon the coexistence of HCl and GSH. The "turn-off" result was attributed to two factors: PH and reducibility. The Gd-/Eu-DPA@Lap could become a promising bimodal probe for biological applications owing to their simple preparation procedure, low toxicity, and high sensitivity.
