囊泡表面的DNA纳米组装体结构调控研究

【摘要】 DNA作为一种遗传信息物质,不仅在基因工程领域受到广泛的关注,更由于其可编程性、可预测性的特点以及功能核酸的发现,使得DNA纳米拓扑结构组装体可用于构造多种分子器件和功能化结构单元[1]。研究DNA纳米组装体与无机纳米材料、高分子聚合物、生物大分子和细胞等相互作用己成为近年来的研究热点之一[2]。本文报道了一项基于囊泡表面的DNA组装体结构调控研究,研究以DNA自组装构建的DNA三棱柱为三维结构模型,通过链置换反应实现了在这种囊泡表面上调控DNA三棱柱聚集和分离。由于囊泡的尺寸可与细胞相媲美,其腔室具有足够的空间可容纳反应分子。另外,基于囊泡的可修饰性特点,该研究可望在调控生物功能分子、构建功能区域和人工细胞中得到广泛应用。

【Abstract】 DNA nanostructure as an idea scaffold has been rationally construct into various architectures for nanotechnology and engineering.Although many of these functionalized DNA assemblies in different settings have been studied ranging from the test tube,cellular conditions to comprehensive surfaces,the behavior on the surface of vesicles has been hardly studied.In the present study,we for the first time report mediating the DNA assemblies on cell-mimicking vesicles including dimerization and disassociation.Here,these kind of vesicles we obtained called Giant Cell Membrane Vesicles(GCMVs) which derived from cancer cells,then DNA triangular prism was used as a 3D-DNA nanostructure model which are DNA-minimal,mediated on GCMVs by DNA strand displacement.Due to properties of GCMVs are similar to cells and which providing a huge space for biochemical reactions,this study may suggest the longer-term feasibility of mediating functional biomoleculars,building delivery system and biomimics.
