活细胞线粒体内膜腺嘌呤核苷转位酶荧光标记的新型方酸菁染料
A Novel Squaraine Dye Targeting for Adenine Nucleotide Translocatase in Inner Mitochondrial Membrane of Live Cells

【摘要】 腺嘌呤核苷转位酶(ANT)是线粒体内外膜上通透性转换孔复合体(PTPC)的重要组成之一,主要分布于细胞线粒体内膜上,在线粒体与细胞质之间的ATP与ADP、AMP及磷酸根离子转运交换过程中发挥重要的作用~1。在一些肿瘤细胞中,异构体(ANT2)表达量会大幅度增加,研究其分布与功能有助于对肿瘤的诊断与治疗。~2本工作中设计合成了一种中位取代新型的方酸菁染料~3,可荧光标记ANT。当染料与MCF-7细胞共培养后,染料分子主要定位于活细胞线粒体内内膜上(Fig.1),线粒体膜上荧光强度比线粒体的基质荧光亮度高,可推测染料主要集中于线粒体膜上;ANT的核心结构上的临位二半胱氨酸残基可与探针反应,进一步的结构证明与功能研究工作尚在进行中。
【Abstract】 Adenine nucleotide translocatase plays a key role in the exchanges of ATP and ADP,AMP and Pi ions between mitochondria matrix and cytosol.In some cancer cells,the up-regulated expression of ANT2 is markedly observed to facilitate tumor disgosis and theropy.We herein designed a new squariane dye based fluorescent tag,composed of a new squaraine dye and a vicinal dithiol binding group.It was found to be mainly localized in the mitochondrial inner membrane upon the incubating with MCF-7 cells,which exhibited brighter fluorescnece emission from the membrane than that of mitochondrial matrix.And there are two cysteine residues in the core structure of ANT to enbale covalently binding to the probe,which further identification and functional exploration is in progress in our group.
