基于适配体-NMOF的电化学法同时检测食品抗生素残留
An Electrochemical Assay for Simultaneous Detection of Several Antibiotics in Foodstuff using Aptamer- Nanoscale MOF

【摘要】 本文开发了一种基于适配体-NMOF的新型多路电化学传感器用于同时检测食品中的多种抗生素残留物,如卡那霉素(KANA),氯霉素(CAP)等。为构建该筛选体系,我们首先合成了具有高比表面多孔NMOF材料(UiO-66-NH2)用于封装大量的金属离子(Pb2+或Cd2+)以及KANA或者CAP的适配体互补链(其互补链分别为cDNA1,cDNA2),该纳米示踪物用NMOF@Mn+/cDNA表示。其次.,KANA和CAP的适配体(分别为Apt1和Apt2)同时标记在磁珠上,形成了识别探针。通过NMOF@Mn+/cDNA上的cDNA和识别探针上的适配体互补杂交,我们得到复合生物编码探针。当适配体特异性识别目标物(KANA和CAP)时,含有Pb2+或Cd2+的纳米示踪物从复合探针中释放。经过磁性分离后,可通过方波伏安法在上清液检测到电化学信号,该信号与目标物的浓度呈线性正相关。该电化学法能显著放大电化学检测信号,所得到的KANA和CAP检测限分别为0.46 pM和0.39 pM(S/N=3),具有高选择性和灵敏性。
【Abstract】 A novel electrochemical assay based on aptamer- NMOF biocodes was developed to monitor several antibiotics including Kanamycin(KANA) and chloramphenicol(CAP) simultaneously.Firstly,the porous amine-functionalized NMOF(UiO-66-NH2) encapsulated with metal ions(Pb2+ or Cd2+) and the complementary strands of aptamers towards KANA and CAP(cDNAl and cDNA2) respectively.Secondly,aptamers of KANA and CAP(Apt1 and Apt2) were simultaneously labeled on magnetic Dynabead as recognition probe.Thirdly,the biocodes were synthesized via hybridization between aptamers with cDNAs.Upon the aptamers bind to the targets(KANA and CAP) specifically,the Pb and Cd ions encoded nanotracers can be released into supernatant after magnetic separation,which be detected by square wave voltammetry(SWV).The assay exhibits high selectivity and sensitivity with detection limits of 0.46 pM and 0.39 pM(S/N=3) towards KANA and CAP respectively.
